植物源复合不育剂ND-2的配比及其对昆明小鼠的不育效果OA
Ratio of plant-derived compound sterility agent ND-2 and its antifertility effect on Kunming mice
不育控制技术是草原鼠害防控的一种有效手段,对实现草原鼠害的可持续控制具有重要意义.本研究通过查阅中国药用植物古籍筛选出 7 种具有潜在抗生育作用的药用植物,以昆明小鼠(Mus musculus)为研究对象,进行不育控制试验.结果表明,蓖麻碱与芍药苷分别对雌性、雄性昆明小鼠具有良好抗生育效果.通过梯度筛选与配方优化后配比的不育剂 ND-2(农大-2 号),对雌雄昆明小鼠不育效果显著:雌性处理组与对照组(仅灌胃 0.2 mL 食用油)相比表现为子宫肿胀导致子宫重量显著增加(P<0.05),繁殖启动期显著提前(P<0.05),较对照组提前 7 d,幼崽成活率显著降低(P<0.05),较对照组降低 5.9%;雄性处理组精子密度和胎仔数显著降低(P<0.05),胎仔数较对照组降低6.8 个,繁殖启动期显著延后(P<0.05),较对照组延后 5 d;雌雄处理组胎仔数(P<0.05),较对照组降低 7.7 个,幼崽成活率显著降低(P<0.05),较对照组降低 7.5%,繁殖启动期显著延后(P<0.05),较对照组延后 4 d.经不育剂 ND-2处理后,雄鼠卵泡刺激素(FSH)、促黄体生成素(LH)、睾酮(T)激素水平显著低于对照组(P<0.05),皮质酮激素水平较对照组出现显著上升(P<0.05);雌鼠激素水平均显著高于对照组(P<0.05).组织学结果显示,雄鼠睾丸间质细胞减少,长形精子形成受限,雌鼠卵巢黄体数量增多、子宫内膜上皮细胞出现变形、死亡.植物源复合不育剂 ND-2 对两性均起到了良好的不育作用,为草原鼠害种群增长的控制提供了新的选择.
The sterile control technique is considered a potential tool for preventing and managing grassland rodent pests,and it holds great significance for achieving sustainable control.In this study,seven medicinal plants with potential antifertility effects were screened out by reviewing ancient Chinese medicinal texts,and sterility control experiments were carried out using Kunming mice as research subjects.The results showed that ricinine and paeoniflorin exhibited strong antifertility effects on female and male Kunming mice,respectively.The sterility agent ND-2(Nongda-2),developed through gradient screening and formulation optimization,demonstrated significant sterility effects in both male and female Kunming mice.Compared with the control group(gavaged with 0.2 mL of edible oil),the female treatment group showed a significant increase in uterine weight due to swelling(P<0.05),an earlier onset of reproductive initiation by 7 days(P<0.05),and a 5.9%reduction in pup survival rate(P<0.05).In the male treatment group,sperm density and litter size were significantly lower than those of the control group(P<0.05);litter size was reduced by 6.8,and reproductive initiation was delayed by 5 days(P<0.05).Across both sexes,litter size was reduced by 7.7(P<0.05),pup survival decreased by 7.5%(P<0.05),and breeding initiation was delayed by 4 days compared with the control group(P<0.05).Hormone assays indicated that,after ND-2 treatment,levels of follicle-stimulating hormone(FSH),luteinizing hormone(LH),and testosterone(T)in male rats were significantly lower than those in the control group(P<0.05),while corticosterone levels were significantly higher(P<0.05).In females,all hormone levels measured were significantly higher than those of the control group(P<0.05).Histological analysis showed that treated males had reduced testicular mesenchymal cells and limited formation of mature spermatozoa,while treated females exhibited an increased number of ovarian corpora lutea along with deformation and death of endometrial epithelial cells.These findings indicate that the plant-derived compound ND-2 has strong sterility effects in both male and female Kunming mice,offering a new option for the control of the growth of grassland rat infestation populations.
