基于网络药理学探讨山芋丸治疗肌少症的作用机制OA

目的:利用网络药理学方法探讨山芋丸治疗肌少症的作用机制.方法:利用TCMSP和BATMAN-TCM数据库,筛选山芋丸的有效成分及其作用靶点;利用GeneCard、DisGeNET、OMIM、CTD数据库综合筛选肌少症疾病靶点;利用Venn图工具获取山芋丸-肌少症交集靶点;采用STRING在线数据库构建交集靶点蛋白相互作用网络,并利用Cytoscape 3.10.0中的CytoHubba插件筛选MCC评分前10的基因作为核心靶点;使用Metascape数据库对交集靶点进行GO功能富集分析和KEGG通路富集分析;最后利用Cytoscape 3.10.0构建"山芋丸-活性成分-靶点-肌少症"可视化网络,并根据网络节点中的度值筛选出山芋丸中排名前10的药物活性成分.结果:共获取山芋丸的活性成分156个,活性成分对应的作用靶点1742个,肌少症疾病靶点2534个,二者的交集靶点121个;排名前十的活性成分中对骨骼肌有积极作用的活性成分为三磷酸腺苷、豆甾醇、棕榈油酸、多巴胺和山奈酚;核心靶点是AKT1、IL6、TNF、IGF1、HIF1A;GO功能富集结果显示:生物过程(Biological Process,BP)、细胞组分(Cellular Component,CC)和分子功能(Molecular Functions,MF),分别获得310、91、108条显著条目,其中BP主要涉及小分子代谢过程的调节,对激素、细胞外刺激、氧化水平的反应等;CC主要涉及膜筏、囊泡腔、细胞皮层、常染色质、刷状缘、核苷酸活化蛋白激酶复合物等;MF主要涉及信号转导受体调节活性、蛋白质同源二聚化活性、转录因子结合、类固醇结合、维生素D结合等.KEGG通路富集分析共获取160个显著条目,参与调控的通路主要为长寿调节通路、精氨酸和脯氨酸代谢、钙信号通路等.结论:山芋丸可能通过多靶点多途径调节骨骼肌蛋白质代谢、线粒体功能等治疗肌少症.

Objective:To explore the mechanism of Shanyu Pills in the treatment of sarcopenia based on network pharmacology.Methods:The active components and corresponding targets of Shanyu Pills were screened from the TCMSP and BATMAN-TCM databases.Sarcopenia-related targets were collected from the GeneCards,DisGeNET,OMIM,and CTD databases.The overlapping targets between Shanyu Pills and sarcopenia were obtained using a Venn diagram tool.A protein-protein interaction(PPI)network of the overlapping targets was constructed using the STRING database,and the CytoHubba plugin in Cytoscape 3.10.0 was used to screen the top 10 genes with the highest MCC scores as key therapeutic targets.Gene Ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed on the overlapping targets using the Metascape database.Finally,a visual"Shanyu Pills-Active Components-Targets-Sarcopenia"network was constructed using Cytoscape 3.10.0,and the top 10 active components of Shanyu Pills were screened according to node degree.Results:A total of 156 active components were obtained from Shanyu Pills,corresponding to 1742 potential targets;2534 sarcopenia-related targets and 121 overlapping targets were identified.Among the top 10 active components,adenosine triphosphate,stigmasterol,palmitoleic acid,dopamine,and kaempferol showed positive effects on skeletal muscle.The core targets included AKT1,IL6,TNF,IGF1,and HIF1A.GO enrichment analysis identified 310 biological processes(BP),91 cellular components(CC),and 108 molecular functions(MF).The BP terms were mainly enriched in the regulation of small molecule metabolic processes,response to hormones,response to extracellular stimuli,and regulation of oxidative stress.The CC terms were mainly enriched in membrane raft,vesicle lumen,cell cortex,euchromatin,brush border,and nucleotide-activated protein kinase complex.The MF terms were mainly enriched in signal transduction receptor activity regulation,protein homodimerization activity,transcription factor binding,steroid binding,and vitamin D binding.KEGG enrichment identified 160 significantly enriched pathways,mainly including the longevity regulating pathway,arginine and proline metabolism,and calcium signaling pathway.Conclusion:Shanyu Pills may exert therapeutic effects on sarcopenia by regulating skeletal muscle protein metabolism and mitochondrial function through multiple targets and pathways.
