电针通过调控IL-33/ST2信号通路抑制缺血性脑卒中大鼠小胶质细胞活化OA
Electroacupuncture inhibits microglial activation in rats with ischemic stroke by regulating the IL-33/ST2 signaling pathway
目的 基于白细胞介素(interleukin,IL)-33/肿瘤发生抑制蛋白2(suppression of tumorigenicity 2,ST2)信号通路对小胶质细胞活化的动态调节作用,探讨电针对缺血性脑卒中大鼠运动功能恢复的潜在机制.方法 将SD大鼠随机分为假手术组、模型组和电针组,用线栓法制备右侧大脑中动脉闭塞模型,再按干预时程分为1、3、7 d 3个亚组.电针选穴为"百会""大椎"及双侧"内关",采用疏密波(5 Hz/10 Hz,1 mA),每日治疗1次,每次20 min.采用改良神经功能缺损评分(modified neurological severity score,mNSS)评估大鼠神经功能;2,3,5-氯化三苯基四氮染色法检测脑梗死体积百分比;苏木精—伊红染色观察缺血区海马组织病理变化;免疫荧光染色法检测缺血皮质区小胶质细胞标志物离子钙接头蛋白1(ionized calcium-binding adaptor molecule 1,Iba1)阳性细胞数;ELISA法检测血清中肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、IL-1β 和 IL-10 表达水平;Western blot 法检测 IL-33与ST2蛋白的表达水平.结果 与假手术组比较,模型组大鼠mNSS评分及脑梗死体积百分比显著升高(P<0.05);海马CA1区神经元排列紊乱,细胞间质水肿导致间隙增宽;Iba1阳性细胞数显著增多(P<0.05);血清TNF-α、IL-1β水平均显著升高(P<0.05),IL-10水平显著降低(P<0.05);IL-33、ST2蛋白表达水平显著升高(P<0.05).与模型组比较,电针组大鼠mNSS评分及脑梗死体积百分比均显著降低(P<0.05);海马CA1区神经元结构完整性显著改善,细胞间质间隙变窄;Iba1阳性细胞数显著减少(P<0.05);血清IL-10水平显著升高(P<0.05),IL-1β、TNF-α水平显著降低(P<0.05);IL-33、ST2蛋白表达水平显著升高(P<0.05).结论 电针可能通过调控IL-33/ST2信号通路,抑制小胶质细胞过度活化,发挥对缺血性脑卒中大鼠的神经保护作用.
Objective To investigate the potential mechanism of electroacupuncture(EA)in promoting motor function recov-ery in rats with ischemic stroke based on the its role in the dynamic regulation of microglial cell activation via the interleukin-33(IL-33)/suppression of tumorigenicity 2(ST2)signaling pathway.Methods Sprague-Dawley rats were randomly divided into sham-operation group,model group,and EA group.The suture method was used to establish a model of right middle cerebral artery occlusion,and then the rats were divided into 1-,3-,and 7-day subgroups based on the duration of intervention.EA was performed at Baihui,Dazhui,and bilateral Neiguan points with a dilatational wave(5 Hz/10 Hz,1 mA)for 20 minutes each time,once a day.Modified Neurological Severity Score(mNSS)was used to assess the neurological function of rats;TTC stai-ning was used to measure the percentage of cerebral infarct volume;HE staining was used to observe the pathological changes of hippocampal tissue;immunofluorescence assay was used to measure the cells positive for ionized calcium-binding adaptor molecule 1(Iba1),a marker for microglial cells,in the ischemic cortex;ELISA was used to measure the serum levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and interleukin-10(IL-10);Western blot was used to measure the protein expression levels of ST2 and IL-33.Results Compared with the sham-operation group,the model group had significant increa-ses in mNSS score and the percentage of cerebral infarct volume(P<0.05),with disordered arrangement of neurons in the hippocampal CA1 region and widened intercellular space due to interstitial edema,as well as a significant increase in the number of Iba1-positive cells(P<0.05),significant increases in the serum levels of TNF-α and IL-1β(P<0.05),a significant reduc-tion in the serum level of IL-10(P<0.05),and significant increases in the protein expression levels of ST2 and IL-33(P<0.05).Compared with the model group,the EA group had significant reductions in mNSS score and the percentage of cerebral infarct volume(P<0.05),with a significant improvement in the structural integrity of neurons in the hippocampal CA1 region and narrowing of intercellular space,as well as a significant reduction in the number of Iba1-positive cells(P<0.05),signifi-cant increases in the serum levels of IL-1β and TNF-α(P<0.05),significant reductions in the levels of IL-1β and TNF-α(P<0.05),and significant increases in the protein expression levels of IL-33 and ST2(P<0.05).Conclusion EA may exert a neuroprotective effect on rats with ischemic stroke by regulating the IL-33/ST2 signaling pathway and inhibiting the overactiva-tion of microglial cells.
