Effect of Electroacupuncture at Neiguan Point on Anxiety-like and Fear-like Behaviors and Neu-ronal Activity in the Prelimbic Cortex in Mice with Post Traumatic Stress Disorder

目的 观察电针"内关"穴对创伤后应激障碍(post traumatic stress disorder,PTSD)小鼠焦虑恐惧样行为、前边缘皮质(prelimbic cortex,PrL)中神经元活动及皮质醇(cortisol,CORT)水平的影响,探讨电针"内关"穴改善PTSD的可能机制.方法 将C57BL/6J小鼠随机分为正常组、模型组、内关组,每组12只.用单一连续刺激加足底电击法复制PTSD小鼠模型.内关组小鼠电针"内关"穴,每日1次,连续14d.采用旷场、高架十字迷宫以及条件恐惧测试实验评估小鼠焦虑、恐惧样行为;光纤记录观察小鼠PrL的神经元活动;酶联免疫吸附试验检测小鼠血清和PrL中CORT水平.结果 与正常组比较,模型组小鼠旷场中央场运动时间、运动距离及跨格次数均显著减少(P<0.05),总静止时间增加(P<0.05);开臂停留时间和进入开臂次数均显著减少(P<0.05);恐惧凝滞时间显著增加(P<0.05);PrL神经元活动显著增加(P<0.05);血清CORT水平显著降低(P<0.05),PrL中CORT水平显著升高(P<0.05).与模型组比较,电针14 d后,内关组小鼠旷场中央场运动时间、运动距离及跨格次数均显著增加(P<0.05),总静止时间显著减少(P<0.05);开臂停留时间和进入开臂次数均显著增加(P<0.05);恐惧凝滞时间显著减少(P<0.05);PrL神经元活动降低(P<0.05),Pearson分析显示,凝滞时间与PrL中神经元活动呈正相关(P<0.05);血清CORT水平显著升高(P<0.05),PrL中CORT水平显著降低(P<0.05).结论 电针"内关"穴可以改善PTSD小鼠焦虑恐惧样行为,其机制可能与抑制PrL神经元活动,调节CORT水平有关.

Objective To investigate the possible mechanism of electroacupuncture at Neiguan point in improving post traumat-ic stress disorder(PTSD)by observing its effect on anxiety-like and fear-like behaviors,neuronal activity in the prelimbic cor-tex,and cortisol(CORT)level in mice with PTSD.Methods C57BL/6J mice were randomly divided into normal group,mod-el group,and Neiguan group,with 12 mice in each group.The method of single prolonged stress and foot shock was used to establish a mouse model of PTSD.The mice in the Neiguan group were given electroacupuncture at Neiguan point once a day for 14 consecutive days.The open field test,the elevated plus maze test,and the fear conditioning test were used to assess the anxiety-like and fear-like behaviors of mice,optical fibers were used to observe neuronal activity in the prelimbic cortex,and ELISA was used to measure the level of CORT in serum and the prelimbic cortex.Results Compared with the normal group,the model group had significant reductions in the time and distance in the central area and the number of squares crossed and a significant increase in total time spent immobile in the open field test(P<0.05),as well as significant reductions in the total time spent in the open arm and the number of open arm entries in the elevated plus maze test(P<0.05)and a significant in-crease in freezing time in the fear conditioning test(P<0.05);compared with the normal group,the model group had a signifi-cant increase in neuronal activity in the prelimbic cortex(P<0.05),a significant reduction in the serum level of CORT(P<0.05),and a significant increase in the level of CORT in the prelimbic cortex(P<0.05).Compared with the model group,the Neiguan group,after 14 days of electroacupuncture,had significant increases in the time and distance in the central area and the number of squares crossed(P<0.05)and a significant reduction in total time spent immobile in the open field test(P<0.05),as well as significant increases in the total time spent in the open arm and the number of open arm entries in the elevated plus maze test(P<0.05),a significant reduction in freezing time in the fear conditioning test(P<0.05),a significant reduction in neuronal activity in the prelimbic cortex(P<0.05),a significant increase in the serum level of CORT(P<0.05),and a signif-icant reduction in the level of CORT in the prelimbic cortex(P<0.05).The Pearson correlation analysis showed that freezing time was positively correlated with neuronal activity in the prelimbic cortex(P<0.05).Conclusion Electroacupuncture at Neiguan point can improve anxiety-like and fear-like behaviors in mice with PTSD,possibly by inhibiting neuronal activity in the prelimbic cortex and regulating the level of CORT.
