水蛭素对糖尿病足溃疡大鼠创面愈合及血管生成的影响OA
Effects of Hirudin on Wound Healing and Angiogenesis of Diabetic Foot Ulcer Rats
目的:探讨水蛭素调控血管内皮生长因子(VEGF)/缺刻基因 1(Notch1)通路对糖尿病足溃疡(DFU)大鼠创面愈合及血管生成的影响.方法:将糖尿病足溃疡大鼠随机分为对照组、模型组、水蛭素低/高剂量组、抑制剂组,每组各 10 只.测定各组大鼠创面愈合率;ELISA法检测血清中 TNF-α、IL-8、IL-6 的表达;HE染色检测大鼠创面组织病理变化;免疫组化染色检测创面组织 CD34 表达和微血管密度(MVD);Western Blot法检测大鼠创面组织中 VEGF、Notch1 蛋白的表达.结果:对照组皮肤组织结构正常,无病理性改变;模型组新生血管分布较少,可见大量炎性细胞浸润;水蛭素低/高剂量组新生血管依次增多,炎性细胞浸润依次减少;抑制剂组新生血管进一步减少,炎性细胞浸润进一步增多.模型组TNF-α、IL-8、IL-6 高于对照组,创面愈合率、CD34 阳性率、微血管密度、VEGF、Notch1 蛋白表达低于对照组,差异有统计学意义(P<0.05);水蛭素低/高剂量组TNF-α、IL-8、IL-6 低于模型组,创面愈合率、CD34 阳性率、微血管密度、VEGF、Notch1 蛋白表达高于模型组,差异有统计学意义(P<0.05);抑制剂组 TNF-α、IL-8、IL-6 高于水蛭素高剂量组,创面愈合率、CD34 阳性率、微血管密度、VEGF、Notch1 蛋白表达低于水蛭素高剂量组,差异有统计学意义(P<0.05).结论:水蛭素可能通过激活VEGF/Notch1 通路,促进糖尿病足溃疡大鼠的创面愈合及血管生成.
Objective:To investigate the effects of hirudin on wound healing and angiogenesis in diabetic foot ulcer(DFU)rats through regulation of the vascular endothelial growth factor(VEGF)/Notch1 pathway.Methods:The DFU rats were randomly divided into the following groups:control group,model group,low-dose hirudin group,high-dose hirudin group,and inhibitor group,with 10 rats in each group.The wound healing rate of rats in each group was measured.ELISA was performed to measure the expression levels of TNF-α,IL-8,and IL-6 in serum.HE staining was performed to assess path-ological changes in rat wound tissue.Immunohistochemical staining was performed to detect CD34 expression and micro-vascular density(MVD)in wound tissue.Moreover,Western Blot was used to measure the expression levels of VEGF and Notch1 proteins in rat wound tissues.Results:The skin tissue structure of the control group was normal and showed no pathological changes.The distribution of new blood vessels in the model group was relatively sparse,and with a large amount of inflammatory cell infiltration.In the low-dose and high-dose thrombin groups,the number of new blood vessels increased successively,while the inflammatory cell infiltration decreased successively.The inhibitor group showed a fur-ther reduction in neovascularization and an increase in inflammatory cell infiltration.The model group had higher levels of TNF-α,IL-8,and IL-6,and lower wound healing rate,CD34 positivity rate,MVD,expression of VEGF and Notch1 than the control group(P<0.05).The low-dose hirudin and high-dose hirudin groups had lower TNF-α,IL-8,and IL-6,and higher wound healing rates,CD34 positivity rates,MVD,VEGF,and Notch1 than the model group(P<0.05).The inhibitor group had higher levels of TNF-α,IL-8,and IL-6,and lower wound healing rate,CD34 positivity rate,MVD,expression of VEGF and Notch1 compared to the high-dose hirudin group(P<0.05).Conclusion:Hirudin may promote wound healing and angiogenesis in DFU rats by activating the VEGF/Notch1 pathway.
