骨碎补总黄酮调控miR-195-5p干预TGF-β1对大鼠生长板软骨细胞增殖的影响OA
Effect of Total Flavonoids of Rhizoma Drynariae on Rat Growth Plate Chondrocyte Proliferation by Regulating miR-195-5p and Intervening in TGF-β1 Action
目的 探讨骨碎补总黄酮通过调控miR-195-5p对大鼠生长板软骨细胞中Ihh-PTHrP通路上TGF-β1表达的影响及其对细胞增殖作用的分子机制.方法 取2周龄SD大鼠胫骨近端生长板软骨细胞进行培养与鉴定,随后采用不同浓度的骨碎补总黄酮(5、20、100、250、500 mg/L)进行体外干预,采用MTT法筛选最佳药物浓度;采用RT-qPCR技术筛选抑制PTHrP表达的质粒;利用MTT法和HE染色法评估骨碎补总黄酮及PTHrP对细胞增殖的影响;ELISA法测定TGF-β1的表达水平.此外,通过生物信息学及双荧光素酶报告基因实验,验证miR-195-5p的靶基因.结果 250 mg/L浓度的骨碎补总黄酮对促进细胞增殖效果最佳,骨碎补总黄酮及PTHrP均能显著提升细胞增殖,而PTHrP抑制剂则具有相反效果.同时验证了miR-195-5p的表达差异及其对TGF-β1表达的调控作用.结论 骨碎补总黄酮可能通过降低miR-195-5p的表达水平,进而提高TGF-β1蛋白的表达,促进生长板软骨细胞的增殖.
Objective To investigate the molecular mechanism by which total flavonoids of rhizoma drynariae affect the proliferation of growth plate chondrocytes in rats,specifically focusing on their effect on TGF-β1 expression in the Ihh-PTHrP pathway via modulating miR-195-5p.Methods Chondrocytes were harvested from the proximal tibial growth plate of two-week-old SD rats,then cultured and identified.Subsequently,in vitro intervention was performed using different concentrations of total flavonoids of rhizoma drynariae(5,20,100,250,500 mg/L).The selection of the optimal drug concentration was carried out using the MTT method,and the screening of plasmids inhibiting PTHrP expression was done by qRT-PCR technology.MTT method and HE staining were employed to evaluate the effect of total flavonoids of rhizoma drynariae and PTHrP on cell proliferation.The expression level of TGF-β1 was determined by ELISA.The target genes of miR-195-5p were validated through bioinformatics analysis and dual-luciferase reporter gene assays.Results The results showed that a concentration of 250 mg/L of total flavonoids of rhizoma drynariae had the best effect on promoting cell proliferation.Both total flavonoids of rhizoma drynariae and PTHrP significantly enhanced cell proliferation,while PTHrP inhibitors had the opposite effect.The differential expression of miR-195-5p and its regulatory role on the expression of TGF-β1 were verified.Conclusion Total flavonoids of rhizoma drynariaemaypromote the proliferation of growth plate chondrocytes by downregulating the expression of miR-195-5p,thereby upregulating TGF-β1 protein expression.
