原花青素B2对地塞米松诱导骨髓间充质干细胞成骨分化的影响OA
Procyanidin B2 on the Mechanism of Osteogenic Differentiation of Dexamethasone-Induced Bone Marrow Mesenchymal Stem Cells
目的:探究原花青素B2 调节 Hippo/Yes相关蛋白(YAP)通路对地塞米松诱导骨髓间充质干细胞(BMSCs)成骨分化的影响.方法:从大鼠体内分离骨髓间充质干细胞,经鉴定后体外培养,分为对照组、地塞米松组、地塞米松+低/中/高剂量原花青素B2 组、地塞米松+高剂量原花青素B2+YAP 激活剂PY-60(地塞米松+高剂量原花青素 B2+PY-60)组.MTT 法检测细胞活力;碱性磷酸酶(ALP)检测试剂盒检测ALP 浓度;茜红素染色检测细胞矿化结节;Western Blot 法检测细胞 Runx2、OCN、Collagen Ⅰ和 Hippo/YAP通路相关蛋白表达.结果:成功分离并完成骨髓间充质干细胞培养.与对照组比,地塞米松组矿化结节数量、OD值、ALP浓度、Runx2、OCN、Collagen Ⅰ蛋白表达降低,p-YAP/YAP比值升高,差异有统计学意义(P<0.05);与地塞米松组比,地塞米松+低/中/高剂量原花青素B2 组矿化结节数量、OD 值、ALP 浓度、Runx2、OCN、Collagen Ⅰ蛋白表达升高,p-YAP/YAP 比值降低,差异有统计学意义(P<0.05);与地塞米松+高剂量原花青素B2 组比,地塞米松+高剂量原花青素B2+PY-60 组矿化结节数量、OD值、ALP 浓度、Runx2、OCN、Collagen Ⅰ蛋白表达降低,p-YAP/YAP比值升高,差异有统计学意义(P<0.05).结论:原花青素 B2 通过抑制 Hippo/YAP 通路减轻地塞米松诱导的骨髓间充质干细胞成骨分化抑制.
Objective:To investigate the effect of procyanidin B2 on dexamethasone-induced osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs)via adjusting the Hippo/Yes-associated protein(YAP)pathway.Meth-ods:BMSCs cells were isolated from rats,identified and cultured in vitro.The cells were then divided into control group,dexam-ethasone group,dexamethasone+low,medium,and high dose procyanidin B2 groups,and dexamethasone+high dose procyanidin B2+YAP activator PY-60(dexamethasone+high dose procyanidin B2+PY-60)group.MTT assay was performed to detect cell viability.The ALP detection kit was used to measure ALP activity.Alizarin Red S staining was performed to visualize and quantify mineralized nodules in cells.Moreover,Western Blot was used to measure the expression levels of Runx2,OCN,Collagen Ⅰ,and Hippo/YAP pathway-related proteins in cells.Results:BMSCs were successfully isolated and cultured.Compared with the control group,the dexamethasone group showed a decreased number of mineralized nodules,OD value,ALP concentration,Runx2,OCN,and Collagen Ⅰ protein expression,while increased p-YAP/YAP ratio(P<0.05).Compared with the dexamethasone group,the dexamethasone+low,medium,and high dose procyanidin B2 groups showed increased number of mineralized nodules,OD value,ALP concentration,Runx2,OCN,and Collagen Ⅰ protein expression,while decreased p-YAP/YAP ratio(P<0.05).Compared with the dexamethasone+high dose procyanidin B2 group,the dexamethasone+high dose procyanidin B2+PY-60 group showed decreased number of mineralized nodules,OD value,ALP concentration,Runx2,OCN,and Collagen Ⅰ protein expression,while increased p-YAP/YAP ratio(P<0.05).Conclusion:Pro-cyanidin B2 alleviates dexamethasone-induced inhibition of osteo-genic differentiation in BMSCs by inhibiting the Hippo/YAP pathway.
