茯苓拮抗咖啡中枢兴奋作用的研究OA
Study on the Antagonistic Effect of Poria cocos on the Central Nervous System Excitation Induced by Coffee
目的 探讨不同浓度的咖啡对中枢的兴奋作用以及茯苓对咖啡兴奋作用的拮抗作用.方法 野生型Canton S黑腹雌性果蝇分为空白组和0.125%、0.25%、0.5%、1%、2%咖啡组,另设空白组和12、24、48 h组,以全天自主活动次数和全天睡眠时间为指标,考察咖啡对果蝇兴奋作用的量效和时效关系,从而选择最佳咖啡浓度和最佳给药时间.再随机分为空白组、模型组、茯苓组和地西泮组,以睡眠自主活动次数和时间进一步探究茯苓对咖啡诱导果蝇自主活动、睡眠时间、睡眠节律的干预作用.结果 1%咖啡给药12 h可显著减少果蝇睡眠时间,增加自主活动次数(P<0.05,P<0.01).与空白组比较,模型组全天、白天和夜晚自主活动次数明显增加(P<0.01),睡眠时间明显降低(P<0.01,P<0.05);与模型组比较,茯苓和地西泮均可显著减少咖啡引起的果蝇自主活动次数,增加睡眠时间(P<0.01,P<0.05).结论 咖啡在较低浓度时就具有强的中枢兴奋作用,而茯苓可以拮抗咖啡的中枢兴奋作用.
Objective To explore the excitatory effects of different concentrations of coffee on the central nervous system and the antagonistic effects of the Poria cocos on the excitatory effects of coffee.Methods Wild-type Canton S female Drosophila melanogaster were divided into a blank group and 5 coffee groups(the concentrations of coffee were 0.125%,0.25%,0.25%,1%,and 2%respectively).Additionally,another blank group,12,24 and 48 h groups were established separately.Taking total daily autonomous activity and total daily sleep time as metrics to reveal the dose-effect and time-effect relationships of coffee on Drosophila excitation.Thereby the best concentrations of coffee and optimal administration time were selected.New cohorts of Drosophila were chosen and randomly divided into the four groups:blank group,model group,Poria cocos group and diazepam group,to further investigate the intervention of Poria cocos in coffee-induced alterations of Drosophila autonomous activity,sleep time,and sleep rhythm.Results 1%coffee administration for 12 h could significantly reduce the sleep time and increase the number of autonomous activities of Drosophila(P<0.01,P<0.05).Compared with the blank group,the model grow group demonstrated a significant increase in total daily,daytime,and nighttime autonomous activity(P<0.01),a significant decrease in sleep time(P<0.01,P<0.05).Compared with the model group,Poria cocos group and diazepam group significantly reduced coffee-induced autonomous activity and increased total sleep time in Drosophila(P<0.01,P<0.05).Conclusion Coffee has strong central nervous system excitatory effects at relatively lower concentrations,while Poria cocos can antagonize the excitatory effects of coffee.
