ADIPOQ基因多态性与策勒黑羊、哈萨克羊和多浪羊生长性状的关联分析OA
Association analysis of ADIPOQ gene polymorphisms with growth traits in Qira black sheep,Kazakh sheep and Dolan sheep
为探究ADIPOQ基因多态性对新疆3个地方品种绵羊(策勒黑羊、哈萨克羊、多浪羊)生长性状的影响,为新疆绵羊的生长性状分子育种提供新的标记资源.该试验以560只绵羊(策勒黑羊173只、哈萨克羊186只、多浪羊201只)为试验对象,收集3个品种1周岁时体高、体重、体斜长和胸围的表型数据;采用Sanger测序与PCR-SSCP技术检测ADIPOQ基因的SNPs,并与其生长性状进行关联分析.结果发现,在ADIPOQ基因的第一外显子中检测到g.201202805G>T错义突变,该突变存在GG、GT和TT3种基因型,其中,哈萨克羊3种基因型频率分别为:0.34、0.55、0.11;多浪羊分别为:0.36、0.54、0.10;策勒黑羊分别为:0.34、0.57、0.09.在3个品种中GT均为优势基因型,G为优势等位基因.关联分析结果表明,哈萨克羊和多浪羊ADIPOQ基因GT型个体的体重、体斜长、体高、胸围显著高于GG和TT型(P<0.05);策勒黑羊中,GT型个体的体重、体斜长、胸围3个性状显著高于GG和TT型(P<0.05),体高无显著差异(P>0.05).综上,ADIPOQ基因对策勒黑羊、哈萨克羊和多浪羊生长性状调控至关重要,其ADIPOQ(g.201202805G>T)位点可作为评估其生长性状的潜在分子遗传标记.
This study was conducted to investigate the effects of ADIPOQ gene polymorphism on the growth traits of three local sheep breeds in Xinjiang namely Qira Black sheep,Kazakh sheep and Duolang sheep,so as to provide novel genetic marker resources for the molecular breeding of growth traits in Xinjiang sheep.A total of 560 sheep individuals,including 173 Qira Black sheep,186 Kazakh sheep and 201 Duolang sheep,were selected as the research objects.Phenotypic data including body height,body weight,body length and chest circumference of the three sheep breeds at one year of age were collected.Sanger sequencing combined with PCR-SSCP technology was used to detect the single nucleotide polymorphisms(SNPs)of ADIPOQ gene,and the association analysis between these SNPs and growth traits was carried out.The results showed that a missense mutation of g.201202805G>T was detected in the exon 1 of ADIPOQ gene,and this mutation site had three genotypes named GG,GT and TT.The frequence of the three genotypes were 0.34,0.55 and 0.11 in Kazakh sheep,0.36,0.54 and 0.10 in Duolang sheep,and 0.34,0.57 and 0.09 in Qira Black sheep.GT was the dominant genotype and G was the dominant allele in all the three breeds.Association analysis results indicated that in Kazakh sheep and Duolang sheep populations,the body weight,body length,body height and chest circumference of individuals with GT genotype were significantly higher than those with GG and TT genotypes(P<0.05).In Qira Black sheep population,the body weight,body length and chest circumference of individuals with GT genotype were significantly higher than those with GG and TT genotypes(P<0.05),while there was no significant difference in body height among individuals with different genotypes(P>0.05).In conclusion,the ADIPOQ gene plays an important role in regulating the growth traits of Qira Black sheep,Kazakh sheep and Duolang sheep,and its g.201202805G>T mutation site can serve as a potential molecular genetic marker for evaluating the growth traits of these sheep breeds.
