[目的]构建酵母文库和酵母单杂交技术筛选JrAGD3 基因的上游调控因子,为解析JrAGD3 基因介导复幼促进不定根发生的分子机制提供理论基础.[方法]以复幼核桃插穗不定根发生过程中插穗基部组织为材料,通过SMART技术构建酵母文库;并以JrAGD3 基因启动子序列为诱饵,采用酵母单杂交技术筛选其上游调控因子.[结果]JrAGD3 基因启动子区域包含多种不同类型的顺式作用元件,分别与光响应调控、激素响应调控、分生组织表达和昼夜节律调控等有关.构建的cDNA文库库容为 3×106 CFU/mL,总克隆数为1.20×107 CFU,平均插入片段长度约 1 000 bp,重组率 100%.酵母单杂交筛选到 19 个潜在的JrAGD3 基因上游调控因子,其中包括WOX4 和WRKY11 转录因子.[结论]本研究构建了高质量的复幼核桃插穗不定根发生过程的酵母文库,可用于酵母单杂交筛库筛选互作蛋白.以JrAGD3 基因启动子序列为诱饵筛选出了WOX4 和WRKY11 转录因子,初步揭示了JrAGD3 基因可能的转录调控网络.研究成果不仅丰富了核桃不定根发生分子机制的研究,也为优化核桃等重要经济林树种扦插繁殖体系、提高无性繁殖效率提供了新的思路.

[Objective]Upstream regulatory factors of the JrAGD3 gene were screened by constructing a yeast library and using yeast one-hybrid technology,aiming to provide a theoretical basis for elucidating the molecular mechanism by which the JrAGD3 gene mediates rejuvenation and promotes adventitious root formation.[Method]Tissues from rejuvenated walnut cuttings undergoing adventitious root formation were used as materials.A yeast cDNA library was constructed using SMART(Switching mechanism At 5′ end of RNA template)technology.The promoter sequence of the JrAGD3 gene was used as a bait to screen for upstream regulatory factors via the yeast one-hybrid(Y1H)system.[Result]The promoter region of JrAGD3 contains multiple types of cis-acting elements,which are associated with light response regulation,hormone response regulation,meristem expression,and circadian rhythm regulation.The constructed cDNA library had a capacity of 3×106 CFU/mL,with a total clone count of 1.20×107 CFU,an average insert fragment length of approximately 1 000 bp,and a recombination rate of 100%.Through yeast one-hybrid screening,19 potential upstream regulatory factors of JrAGD3 were identified,including two transcription factors:WOX4 and WRKY11.[Conclusion]This study successfully constructed a high-quality yeast library for the rejuvenation-induced adventitious root formation process in walnut cuttings,providing a valuable resource for screening interacting proteins using the yeast one-hybrid system.Using the JrAGD3 gene promoter sequence as bait,WOX4 and WRKY11 transcription factors were identified,preliminarily revealing the potential transcriptional regulatory network of JrAGD3.These findings not only enriched the molecular understanding of adventitious root formation in walnut but also provided new insights for optimizing cutting propagation systems and improving the clonal propagation efficiency of walnut and other important economic forest tree species.
