绿原酸治疗中枢神经系统疾病作用及其机制研究进展OA
Research Progress in Effects and Mechanisms of Chlorogenic Acid in Treating Central Nervous System Diseases
当前中枢神经系统疾病临床治疗药物长期应用易出现疗效减退及不良反应.绿原酸作为天然多酚类化合物,被证实具有显著的中枢神经保护活性.其作用机制呈现多靶点、多通路协同特点,主要包括:抑制炎症信号轴及促炎因子释放,从而发挥抗炎作用;增强抗氧化酶活性,减轻氧化应激损伤;调节突触表达及海马突触可塑性;稳定线粒体功能,抑制神经元凋亡与铁死亡;通过肠-脑轴调控菌群-神经免疫交互网络.对绿原酸在阿尔茨海默病、帕金森病、癫痫、脑卒中、抑郁、焦虑、脊髓损伤、慢性疼痛、创伤后应激障碍治疗中发挥作用的分子机制,以及相关纳米递送技术与联合治疗策略的创新应用进行综述,旨在深入阐明绿原酸的多维神经保护机制,以期为相关药物研发及中枢神经系统疾病的防治提供新思路与理论依据.
Currently,long-term use of clinical therapeutic drugs for central nervous system(CNS)diseases is prone to diminished efficacy and adverse reactions.As a natural polyphenolic compound,chlorogenic acid(CGA)has been proven to possess significant central neuroprotective activity.It exhibits multi-targeted and multi-pathway synergistic therapeutic mechanisms,which mainly include inhibiting inflammatory signaling axes and the release of pro-inflammatory factors to exert anti-inflammatory effects,enhancing antioxidant enzyme activity to alleviate oxidative stress damage,regulating synaptic expression and hippocampal synaptic plasticity,stabilizing mitochondrial function to inhibit neuronal apoptosis and ferroptosis,and regulating the microbiota-neuroimmune crosstalk network via the gut-brain axis.This review summarizes the molecular mechanisms through which CGA exerts therapeutic effects on Alzheimer's disease(AD),Parkinson's disease(PD),epilepsy,stroke,depression,anxiety,spinal cord injury(SCI),chronic pain,and post-traumatic stress disorder(PTSD),as well as the innovative applications of related nano-delivery technologies and combined therapeutic strategies.This review aims to comprehensively clarify the multidimensional neuroprotective mechanisms of CGA,thereby providing new ideas and a theoretical basis for the development of related drugs and the prevention and treatment of CNS diseases.
