目的:建立经典名方桔梗汤的高效液相色谱法(HPLC)特征图谱,并基于一测多评法测定其主要成分的含量.方法:采用Phenomenex-C18 色谱柱(250 mm×4.6 mm,5 μm),以 0.05%磷酸水溶液(A)-乙腈(B)为流动相梯度洗脱,采用二极管阵列检测器,检测波长为 203 nm,柱温为 35℃,流速为 1.0 mL·min-1,建立桔梗汤的HPLC特征图谱,对其共有峰进行指认、归属,并对 15 批桔梗汤的HPLC特征图谱与对照图谱间的相似度进行分析.采用一测多评法测定桔梗汤中去芹糖桔梗皂苷E、桔梗皂苷D、甘草苷、甘草酸 4 个主要成分的含量.结果:建立了桔梗汤的HPLC特征图谱,共标定 14 个特征峰,指认峰 5、峰 9、峰 12、峰 14 代表的化学成分分别为甘草苷、甘草酸、去芹糖桔梗皂苷E、桔梗皂苷D,其中甘草苷、甘草酸来自甘草,芹糖桔梗皂苷E、桔梗皂苷D来自桔梗.15 批桔梗汤HPLC特征图谱与对照图谱间的相似度均大于 0.900.15 批桔梗汤中去芹糖桔梗皂苷E、桔梗皂苷D、甘草苷、甘草酸的质量分数分别为 0.31~0.35、5.43~6.14、6.31~7.05、10.35~12.45 mg·g-1.结论:建立的HPLC特征图谱和一测多评法高效、可行,为经典名方桔梗汤的质量评价提供参考.

Objective:To establish the high-performance liquid chromatography(HPLC)characteristic chromatograms of the classic formula Jiegeng Decoction and determine the content of its major components using the quantitative analysis of multi-components by single marker(QAMS)method.Methods:Separation was conducted on gradient elution through a Phenomenex-C18 column(250 mm×4.6 mm,5 μm)with a mobile phase consisting of 0.05%phosphoric acid aqueous solution(A)and acetonitrile(B)at a column temperature of 35℃and a flow rate of 1.0 mL·min-1.Detection was performed with a diode array detector at a wavelength of 203 nm.The HPLC characteristic chromatograms of Jiegeng Decoction were established,and the common peaks were identified and assigned.The similarity between the HPLC characteristic chromatograms of 15 batches of Jiegeng Decoction and the reference characteristic chromatogram was evaluated.The QAMS method was employed to determine the content of four major components(glycyrrhizin,glycyrrhetic acid,deapioplatycodin E,and platycodin D)in Jiegeng Decoction.Results:The HPLC characteristic chromatograms of Jiegeng Decoction were successfully established,with 14 characteristic peaks identified.The peaks 5,9,12,and 14 were identified as glycyrrhizin,glycyrrhetic acid,deapioplatycodin E,and platycodin D,respectively.Glycyrrhizin and glycyrrhetic acid were derived from Glycyrrhizae Radix et Rhizoma,while deapioplatycodin E and platycodin D were from Platycodonis Radix.Furthermore,the similarity between the characteristic chromatograms of the fifteen batches of Jiegeng Decoction and the reference characteristic chromatogram was greater than 0.900.The contents of deapioplatycodin E,platycodin D,glycyrrhizin,and glycyrrhetic acid in the 15 batches of Jiegeng Decoction were in the ranges of 0.31-0.35,5.43-6.14,6.31-7.05,and 10.35-12.45 mg·g-1,respectively.Conclusion:The established HPLC characteristic chromatograms combined with the QAMS method are efficient and feasible,providing a comprehensive scientific basis for the quality evaluation of the classic formula Jiegeng Decoction.
