川芎中21种元素分析及风险评估OA
Analysis and Risk Assessment of 21 Elements in Ligusticum chuanxiong
目的:对川芎药材、茎叶、根土及灌溉水中 21 种元素含量进行测定,并对药材中有害元素铅(Pb)、镉(Cd)、砷(As)、汞(Hg)、铜(Cu)、铝(Al)、铬(Cr)、镍(Ni)等进行风险评估,以期为川芎的质量安全评价提供依据.方法:样品经微波消解后,采用电感耦合等离子体质谱法(ICP-MS)对川芎中As、Hg、Cu、Pb、Cd、Al、Cr、Ni等 21 种元素进行分析,在此基础上进行健康风险评估.结果:21 种元素的线性关系、精密度良好;检出限为 0.001~0.612 μg·kg-1;回收率为 85%~115%.川芎中钾(K)、钙(Ca)、铁(Fe)、Al、钠(Na)的质量分数均大于 50 mg·kg-1,占比较高.5 批川芎中有害元素危害指数(HI)均小于 1,表明非致癌性风险较低,可被接受;Pb、As的致癌风险(CR)均低于 1×10-4,致癌性风险较低,可被接受.川芎药材中的重金属含量与其茎叶中的重金属含量之间存在显著的正相关关系.结论:建立的川芎多元素分析方法具有较好专属性及灵敏度,结合风险评估技术,可为川芎的质量安全评价提供技术支持.
Objective:To determine the content of 21 elements in Ligusticum chuanxiong herbs,stems,leaves,rhizosphere soil,and irrigation water,and assess the risk of toxic elements in the medicinal material,including Pb,Cd,As,Hg,Cu,Al,Cr,and Ni,aiming to provide a basis for the quality and safety evaluation of L.chuanxiong.Methods:After microwave digestion,inductively coupled plasma mass spectrometry(ICP-MS)was employed to analyze 21 elements.Based on the results,health risks were assessed.Results:The 21 elements exhibited good linearity and precision,with limits of detection ranging from 0.001 to 0.612 μg·kg-1,and recovery rates of 85%-115%.The mass fractions of K,Ca,Fe,Al,and Na in L.chuanxiong were greater than 50 mg·kg-1,accounting for a high proportion.The hazard index(HI)of harmful elements in five batches of samples was less than 1,indicating a low and acceptable non-carcinogenic risk.The carcinogenic risk(CR)of Pb and As was lower than 1×10-4,indicating a low and acceptable carcinogenic risk.There was a significant positive correlation between the heavy metal content in L.chuanxiong herbs and that in the stems and leaves.Conclusion:The multi-element analysis method established in this study demonstrates satisfactory specificity and sensitivity.Combined with risk assessment technology,it can provide technical support for the quality safety evaluation of L.chuanxiong.
