SLC7A5促进TGF-β1诱导的小鼠胚胎成纤维细胞活化和增殖OA
SLC7A5 promotes TGF-β1-induced activation and proliferation of mouse embryonic fibroblasts
目的 探讨溶质载体家族7成员5(SLC7A5)对转化生长因子β1(TGF-β1)诱导的小鼠胚胎成纤维细胞活化和增殖的作用.方法 小鼠胚胎成纤维细胞系NIH-3T3用TGF-β1孵育24 h,使用PCR阵列(PCR array)筛选靶基因.使用沉默慢病毒sh-SLC和重表达慢病毒r-SLC转染成纤维细胞.采用Western blotting和实时定量PCR检测目标靶点SLC7A5以及成纤维细胞活化指标α平滑肌肌动蛋白(α-SMA)、波形蛋白(Vim)和Ⅰ型胶原(COL-Ⅰ)合成;采用CCK-8法检测各组成纤维细胞的增殖速度;采用DCFH-DA探针和还原型谷胱甘肽检测试剂盒检测成纤维细胞内氧化还原状态.结果 与PBS组相比,TGF-β1组成纤维细胞中SLC7A5蛋白和基因表达水平均升高(P<0.05).与未转染病毒对照组(Con组)相比,SLC7A5基因沉默组(sh-SLC组)成纤维细胞中α-SMA、Vim和COL-Ⅰ表达水平下降,增殖速度降低,谷胱甘肽含量减少,活性氧累积加重(P<0.05);与sh-SLC组相比,重新表达SLC7A5基因组(r-SLC组)成纤维细胞中上述现象能够逆转.结论 SLC7A5促进TGF-β1诱导的小鼠胚胎成纤维细胞活化和增殖,并可能通过调节细胞内氧化应激状态发挥作用,SLC7A5有望成为纤维化疾病中抑制成纤维细胞活化和增殖的靶点.
Objective To investigate the effect of solute carrier family 7 member 5(SLC7A5)on the activation and proliferation of mouse embryonic fibroblasts induced by transforming growth factor β1(TGF-β1).Methods The NIH-3T3 mouse embryonic fibro-blast line was incubated with TGF-β1 for 24 h,and the target genes were screened using a PCR array.The fibroblasts were transfected with lentiviral vectors carrying either sh-SLC RNA(for gene silencing)or r-SLC RNA(for gene re-expression).The expression levels of SLC7A5,α-smooth muscle actin(α-SMA),vimentin(Vim),and type Ⅰ collagen(COL-Ⅰ)were detected using Western blotting and real-time PCR.The CCK-8 assay was used to determine the fibroblast proliferation rate in each group.The intracellular reduction-oxidation status was detected using a DCFH-DA probe and a reduced glutathione detection kit.Results The SLC7A5 protein and mRNA expres-sion levels in the TGF-β1 group were higher than those in the PBS group(P<0.05).Moreover,the α-SMA,Vim,and COL-Ⅰ expression levels were lower in the sh-SLC group than in the non-virus-transfected control group(P<0.05).Additionally,the proliferation rate of the sh-SLC-transfected fibroblasts and their intracellular glutathione content were decreased and their accumulation of reactive oxygen spe-cies was aggravated(P<0.05).By contrast,these effects were reversed in the r-SLC group.Conclusion SLC7A5 promotes the TGF-β1-induced activation and proliferation of mouse embryonic fibroblasts and may play a role in regulating intracellular oxidative stress.There-fore,SLC7A5 is a potential target for therapies aimed at inhibiting the activation and proliferation of fibroblasts in fibrotic diseases.
