栀子提取液对急性出血坏死性胰腺炎小鼠炎性介质的影响研究OA
Effects of Gardenia Extract on Inflammatory Mediators in Mice with Acute Hemorrhagic Necrotizing Pancreatitis
目的 观察栀子提取液对急性出血坏死性胰腺炎小鼠炎性介质的影响.方法 将BALB/c小鼠随机分为假手术组、模型组、栀子组、激活剂组,每组8只.制备急性出血坏死性胰腺炎模型,各组给予相应药物治疗.检测小鼠血清中白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、白细胞介素-4(IL-4)、白细胞介素-10(IL-10)水平和淀粉酶、脂肪酶含量;计算胰腺指数;苏木精-伊红(HE)染色检测胰腺组织病理学变化并对损伤进行评分;免疫组化染色检测胰腺组织中髓过氧化物酶(MPO)表达;蛋白质印迹法(Western blotting)检测胰腺组织中p38MAPK、p-p38MAPK、NF-κB p65、p-NF-κB p65蛋白表达.结果 与假手术组相比,模型组小鼠血清中促炎因子TNF-α、IL-1β、IL-6水平、淀粉酶、脂肪酶、胰腺指数、MPO阳性细胞数、胰腺组织中p-p38MAPK、p-NF-κB p65蛋白表达均明显升高,抗炎因子IL-4、IL-10水平均明显降低(P<0.001);与模型组相比,栀子组小鼠血清中促炎因子TNF-α、IL-1β、IL-6水平、淀粉酶、脂肪酶、胰腺指数、MPO阳性细胞数、胰腺组织中p-p38MAPK、p-NF-κB p65蛋白表达均明显降低,抗炎因子IL-4、IL-10水平均明显升高(P<0.001);与栀子组相比,激活剂组小鼠血清中促炎因子TNF-α、IL-1β、IL-6水平、淀粉酶、脂肪酶、胰腺指数、MPO阳性细胞数、胰腺组织中p-p38MAPK、p-NF-κB p65蛋白表达明显升高,抗炎因子IL-4、IL-10水平均明显降低(P<0.001).结论 栀子提取液可抑制急性出血坏死性胰腺炎小鼠炎症反应,改善胰腺组织病理学损伤,其作用机制可能与抑制p38MAPK/NF-κB信号的激活有关.
Objective:To investigate the effects of gardenia extract on inflammatory mediators in mice with acute hemorrhagic necrotizing pancreatitis.Methods:BALB/c mice were randomly divided into the sham opera-tion group,model group,gardenia group and activator group,with 8 mice in each group.The acute hemorrhagic necrotizing pancreatitis model was established,and each group was administered the corresponding drug treatment.The serum levels of IL-6,IL-1β,TNF-α,IL-4,IL-10,amylase and lipase were detected;the pancreatic index was calculated;hematoxylin-eosin(HE)staining was used to observe the pathological changes of pancreatic tissue and score the damage;immunohistochemical staining was applied to detect the expression of myeloperoxidase(MPO)in pancreatic tissue;Western blotting was performed to determine the protein expressions of p38MAPK,p-p38MAPK,NF-κB p65 and p-NF-κB p65 in pancreatic tissue.Results:Compared with the sham operation group,the serum levels of pro-inflammatory factors(TNF-α,IL-1β,IL-6),amylase,lipase,pancreatic index,number of MPO-posi-tive cells,and protein expressions of p-p38MAPK and p-NF-κB p65 in pancreatic tissue of the model group were significantly increased,while the serum levels of anti-inflammatory factors(IL-4,IL-10)were significantly de-creased(P<0.001).Compared with the model group,the serum levels of pro-inflammatory factors(TNF-α,IL-1β,IL-6),amylase,lipase,pancreatic index,number of MPO-positive cells,and protein expressions of p-p38MAPK and p-NF-κB p65 in pancreatic tissue of the gardenia group were significantly decreased,while the serum levels of anti-inflammatory factors(IL-4,IL-10)were significantly increased(P<0.001).Compared with the gardenia group,the serum levels of pro-inflammatory factors(TNF-α,IL-1β,IL-6),amylase,lipase,pancreatic index,num-ber of MPO-positive cells,and protein expressions of p-p38MAPK and p-NF-κB p65 in pancreatic tissue of the activator group were significantly increased,while the serum levels of anti-inflammatory factors(IL-4,IL-10)were significantly decreased(P<0.001).Conclusion:Gardenia extract can inhibit the inflammatory response and improve the pathological damage of pancreatic tissue in mice with acute hemorrhagic necrotizing pancreatitis,and its mechanism may be related to the inhibition of p38MAPK/NF-κB signaling pathway activation.
