目的 探讨天麻素(GAS)对牙周炎大鼠炎性损伤及对AMP依赖的蛋白激酶/核因子κB/核苷酸结合寡聚化结构域样受体蛋白 3(AMPK/NF-κB/NLRP3)通路的影响.方法 构建牙周炎大鼠模型,将造模成功大鼠随机分为模型(Model)组、天麻素低、中、高剂量(GAS-L、GAS-M、GAS-H)组、天麻素高剂量+通路抑制剂Compound C(GAS-H+CC)组,每组18 只,另取18 只健康正常大鼠作为对照(Control)组;评定大鼠牙龈指数和牙龈出血指数;显微计算机断层扫描仪(Micro-CT)测定牙周炎诱导区域釉牙骨质界和牙槽骨嵴顶(CEJ-ABC)距离及骨密度;酶联免疫吸附试验(ELISA)检测血清及牙周组织炎性因子水平;苏木精-伊红(HE)染色检测牙周组织病理损伤;抗酒石酸酸性磷酸酶(TRAP)染色检测牙周组织破骨细胞数量;免疫组化检测牙周组织骨代谢相关蛋白表达;免疫印迹(Westem blot)检测AMPK/NF-κB/NLRP3 通路相关蛋白表达.结果 GAS-L、GAS-M、GAS-H组较Model组牙周组织病理损伤减轻,骨吸收陷窝减少,炎性细胞浸润减轻,牙龈指数、牙龈出血指数、CEJ-ABC距离、肿瘤坏死因子-α(TNF-α)、白介素(IL)-6 水平、组织损伤评分及破骨细胞数量、核因子κB受体活化因子配体(RANKL)、p-NF-κB p65/NF-κB p65、NLRP3 表达降低(P<0.05,P<0.01),骨密度、骨保护素(OPG)、骨钙素(OCN)、p-AMPK/AMPK表达升高(P<0.05,P<0.01);GAS-H+CC组较GAS-H组牙周组织病理损伤加重,牙龈指数、牙龈出血指数、CEJ-ABC距离、TNF-α、IL-6 水平、组织损伤评分及破骨细胞数量、RANKL、p-NF-κB p65/NF-κB p65、NLRP3 表达升高,骨密度、OPG、OCN、p-AMPK/AMPK表达降低(均P<0.05).结论 GAS可改善牙周炎大鼠炎性损伤,与调控AMPK/NF-κB/NLRP3 通路相关.

Objective To investigate the effect of gastrodin(GAS)on inflammatory injury and the AMP-dependent protein kinase/nuclear factor κB/nucleotide-binding oligomerization domain-like receptor protein 3(AMPK/NF-κB/NLRP3)pathway in rats with periodontitis.Methods A periodontitis rats model was constructed,and successfully modeled rats were randomly assigned into either the Model group,low,medium,and high-dose GAS(GAS-L,GAS-M,and GAS-H,respectively),or high-dose GAS+pathway inhibitor compound C group(GAS-H+CC),with 18 rats in each.Additionally,18 healthy normal rats formed the Control group.The gingival index and the gingival bleeding index of rats were evaluated.Microscopic computed tomography was used to measure alveolar bone resorption,cemento-enamel junction-alveolar bone crest(CEJ-ABC)distance,and bone mineral density.Enzyme-linked immunosorbent assay was used to detect levels of inflammatory factors in serum and periodontal tissues.Hematoxylin-eosin staining was performed to detect pathological damage in periodontal tissue.Tartrate-resistant acid phosphatase staining was performed to detect the number of osteoclasts in periodontal tissue.Immunohistochemical detection of the expression of bone metabolism-related proteins in periodontal tissues was performed,and Western blot was used to detect the expression of AMPK/NF-κB/NLRP3 pathway-related proteins.Results Compared with the Model group,the GAS-L,GAS-M,and GAS-H groups showed reduced periodontal tissue pathological damage,bone resorption cavities,inflammatory cell infiltration,gingival index,gingival bleeding index,CEJ-ABC distance,tumor necrosis factor(TNF)-α,interleukin(IL)-6,tissue damage score,osteoclast count,RANKL,p-NF-κB p65/NF-κB p65,and NLRP3,while bone mineral density,osteoprotegerin(OPG),osteocalcin(OCN),and p-AMPK/AMPK increased(P<0.05,P<0.01).The GAS-H+CC group showed more severe periodontal tissue pathological damage than the GAS-H group;the gingival index,gingival bleeding index,CEJ-ABC distance,TNF-α,IL-6,tissue damage score,osteoclast count,RANKL,p-NF-κB p65/NF-κB p65,and NLRP3 increased,while bone mineral density,OPG,OCN,and p-AMPK/AMPK decreased(P<0.05,P<0.01).Conclusions GAS improved inflammatory injury in periodontitis rats through regulating the AMPK/NF-κB/NLRP3 pathway.
