正交试验结合AHP-CRITIC法优化芍药甘草汤水煎液提取工艺OA
Optimization of the extraction process of Shaoyao Gancao decoction by orthogonal test and AHP-CRITIC analyses
采用正交试验结合多指标综合加权评分法优化芍药甘草汤水煎液的提取工艺.建立高效液相色谱(HPLC)同时测定水煎液中芍药苷、芍药内酯苷、甘草苷和甘草酸 4种指标成分含量的方法;选择L9(34)正交表考察浸泡时间、料液比、煎煮时间及提取次数 4种因素及不同水平对芍药甘草汤水提工艺的影响;以出膏率及 4种指标成分质量分数为评价指标,综合采用AHP、CRITIC、AHP-CRITIC混合加权法确定各指标权重系数,结合正交试验对芍药甘草汤水煎液提取工艺进行优选.确定芍药甘草汤的最佳提取工艺为:料液比 1:8 g/mL,浸泡 20 min后提取 3次,每次 60 min.正交试验联合AHP-CRITIC混合加权法为芍药甘草汤水煎液的优化提取提供了一种可靠、稳定的方案.
To optimize the extracting process of Shaoyao Gancao decoction,a multi-index comprehensive weighted scoring method combined with an orthogonal test was applied.An HPLC method was established for the simultaneous determination of 4 ingredients including paeoniflorin,albiflorin,liquiritin and glycyrrhizic acid.The L9(34)orthogonal table was adopted to investigate the effects of soaking time,water amount,decocting time and extracting times on the water-extracting process of Shaoyao Gancao decoction.Yield ratios and the contents of index components were used as evaluation indexes,and the weight coefficient of each index was determined by using AHP,CRITIC,or AHP-CRITIC method.The extracting process parameters of Shaoyao Gancao decoction were then determined by analyses in the orthogonal test.The optimized extraction process of Shaoyao Gancao decoction was confirmed to involve adding an eightfold amount of water(w/v),soaking for 20 minutes,and then extracting three times for 60 minutes each time..The orthogonal test method combined with AHP-CRITIC mixed weighting method provided a reliable and stable scheme for the optimized extracting of Shaoyao Gancao decoction.
