天麻极细粉重复给药毒性试验研究OA
Study on the repeated oral toxicity of Gastrodia elata ultrafine powder
旨在探讨重复给予天麻极细粉对Sprague-Dawley(SD)大鼠的毒性反应,为其药用安全性评价提供科学依据.将 128只SD大鼠随机分为阴性对照组、天麻极细粉低、中、高剂量组(0.7、1.4、2.8 g/kg),每组 32只,雌雄各半.各组大鼠连续给药 26周,给药结束设恢复期 4周,每日观察大鼠的一般状况,每周测定大鼠体重和摄食量,于给药末期和恢复期末进行解剖,测定血液学、血清生化、电解质、凝血、主要脏器系数指标,肉眼及镜下观察组织形态.结果与阴性对照组比较,雌、雄各剂量组大鼠各项指标均未见与受试物相关的毒理学变化.在本试验条件下,2.8 g/kg为最大未观察到天麻极细粉有害作用剂量,日常大量长期食用时需重点观察食欲、体重的变化情况.
This study aimed to investigate the toxic effects of repeated administration of ultrafine powder of Gastrodia elata in Sprague-Dawley(SD)rats and to provide a scientific basis for its safety evaluation in medicinal use.A total of 128 SD rats were randomly divided into a negative control group and low,medium,and high-dose groups of Gastrodia elata ultrafine powder(0.7,1.4,and 2.8 g/kg),with 32 rats in each group and an equal number of males and females.All rats were administered the test substance continuously for 26 weeks,followed by a 4-week recovery period after the end of administration.The general condition of the rats was observed daily,and body weight and food intake were measured weekly.At the end of the dosing period and the recovery period,necropsy was performed,and hematological parameters,serum biochemical indices,electrolytes,coagulation function,and major organ coefficients were determined.Tissue morphology was examined both macroscopically and microscopically.Compared with the negative control group,no toxicological changes related to the test substance were observed in any of the measured parameters in either male or female rats at all dose levels.Under the conditions of this study,2.8 g/kg was identified as the no-observed-adverse-effect level(NOAEL)for Gastrodia elata ultrafine powder.However,during long-term high-dose consumption in daily use,particular attention should be paid to changes in appetite and body weight.
