药用植物雀舌木属叶绿体基因组的比较研究OA
Comparative analysis of the chloroplast genome of the medicinal plant Leptopus Decne.
对雀舌木属具药用价值的雀儿舌头与雀舌木叶绿体基因组比较分析,明确基因序列的特征、密码子使用偏好及系统发育规律.采用二代高通量测序技术对雀儿舌头叶绿体全基因组进行测序、组装与注释,并对基因组结构、重复序列、密码子使用偏好以及系统发育分析.结果表明,雀舌木属叶绿体基因组呈典型双链环状四分体,反向重复区(IR)保守性高,大单拷贝区(LSC)、小单拷贝区(SSC)变异较多,且LSC富含简单重复序列(SSR).密码子使用呈现非中性,第三位碱基偏好A/U.基于叶绿体全基因组构建系统发育树以 100%支持率揭示二者的密切亲缘关系.选择压力在密码子偏好性形成中发挥较大影响,为辅助研究雀舌木属天然成分代谢及利用提供基因数据支持.
To clarify the characteristics of Leptopus Decne.sequences,codon usage patterns,influencing factors,and phylogeny,we perform a comparative analysis of the chloroplast genomes of Leptopus chinensis and L.cordifolius with medicinal value.The whole genome of L.chinensis chloroplasts is sequenced,assembled,and annotated by second-generation high-throughput sequencing technology,and further analyzes its genome structure,repeat sequences,codon usage preference,and phylogeny.The chloroplast genome of Leptopus exhibited a typical double-stranded circular tetrad.The IR region reflects high conservatism,and there are more mutation sites in the LSC and SSC regions,while the LSC region contains more SSRs.Codon usage analysis indicated a non-neutral,with a preference for using A/U at the third codon position.The phylogenetic tree constructed based on the whole genome of chloroplasts revealed a close phylogenetic relationship with 100%suppor.In conclusion,selection pressure has a great influence on the codon preference of the chloroplast genome of Leptopus,and thess results can provide genetic data support for futher studies on the metabolism and utilization of natural components of Leptopus.
