乳腺癌组织中QPRT和SHMT2的表达及其临床意义OA
Expressions of QPRT and SHMT2 in breast cancer tissues and their clinical significance
目的 探讨乳腺癌组织中喹啉酸磷酸核糖转移酶(quinolinate phosphoribosyltransferase,QPRT)和丝氨酸羟甲基转移酶2(serine hydroxymethyltransferase 2,SHMT2)的表达及其与临床病理特征和预后的关系,以及两者的相关性.方法 收集 2020年 1 月至 2023 年 6 月云南省肿瘤医院存档的 236 例非特殊类型乳腺癌和其中 118 例癌旁乳腺组织.采用免疫组织化学法检测组织中QPRT和SHMT2 的蛋白表达.通过Kaplan-Meier plotter数据库分析乳腺癌组织中QPRT和SHMT2 表达对患者预后的影响.利用基因表达谱交互式分析(Gene Expression Profiling Interactive Analysis,GEPIA)数据库分析QPRT和SHMT2 在乳腺癌组织中的mRNA表达及其相关性.结果 236 例乳腺癌组织中QPRT和SHMT2 的高表达率分别为 25.0%和 66.5%,高于癌旁组织的 5.1%和 51.7%(均P<0.01).QPRT表达水平在TNM分期、组织学分级和淋巴结转移方面比较,差异均具有统计学意义(均P<0.05).SHMT2 表达水平在TNM分期和淋巴结转移方面比较,差异均具有统计学意义(均P<0.05).QPRT表达在人表皮生长因子受体 2(human epidermal growth factor receptor 2,HER2)过表达型乳腺癌中高于管腔A型、管腔B1 型和三阴性乳腺癌(均P<0.05).乳腺癌不同分型中SHMT2 表达比较,差异无统计学意义(P>0.05).Kaplan-Meier plotter数据库分析显示,乳腺癌组织中QPRT和SHMT2 低表达患者总生存期、无远处转移生存期和无复发生存期均优于高表达患者,且SHMT2 低表达患者进展后生存期也优于高表达患者(均P<0.05).GEPIA数据库分析显示,乳腺癌组织中QPRT和SHMT2 表达呈正相关(r=0.23,P<0.01).结论 QPRT和SHMT2 高表达均与乳腺癌的发生、发展和不良预后有关.QPRT表达与乳腺癌的不同分型有关.乳腺癌组织中QPRT和SHMT2 表达呈正相关.
Objective To investigate the expressions of quinolinate phosphoribosyltransferase(QPRT)and serine hydroxymethyltrans-ferase 2(SHMT2)in breast cancer tissues and their correlation with the clinicopathological characteristics and prognosis of the patients.Methods A total of 236 cases of non-special type breast carcinoma and 118 matched adjacent normal breast tissues archived in Yunnan Cancer Hospital from January 2020 to June 2023 were enrolled in this study.The expressions of QPRT and SHMT2 proteins in tissues were detected by immunohistochemistry.The Kaplan-Meier plotter database was used to analyze the impact of QPRT and SHMT2 expressions in breast cancer tissues on the prognosis of the patients.The Gene Expression Profiling Interactive Analysis(GEPIA)database was utilized to analyze the mRNA expressions of QPRT and SHMT2 in breast cancer tissues and their correlation.Results The high expression rates of QPRT and SHMT2 in breast cancer tissues were significantly higher than those in adjacent tissues(QPRT:25.0%vs 5.1%,SHMT2:66.5%vs 51.7%,both P<0.01).The expression of QPRT protein in breast cancer tissues was significantly different in terms of TNM stage,histological grade,and lymph node metastasis(all P<0.05),while the expression of SHMT2 protein was significantly different in terms of TNM stage and lymph node metastasis(both P<0.05).QPRT expression in human epidermal growth factor receptor 2(HER2)-overexpressing breast cancer tissues was higher than those in luminal A,luminal B1,and triple-negative breast cancer tissues(all P<0.05).No significant difference in SHMT2 expression was observed among different breast cancer molecular subtypes(P>0.05).The Kaplan-Meier plotter database analysis revealed that breast cancer patients with either low QPRT or low SHMT2 expression exhibited superior overall survival,distant metasta-sis free survival,and recurrence-free survival compared to those with high expression of QPRT or SHMT2(all P<0.05).Furthermore,low SHMT2 expression was also associated with improved post-progression survival(P<0.05).The GEPIA database analysis showed that there was a positive correlation between QPRT and SHMT2 protein expressions in breast cancer tissues(r=0.23,P<0.01).Conclusions The high expressions of QPRT and SHMT2 is related to the occurrence,development,and poor prognosis of breast cancer.The expression of QPRT is related to different types of breast cancer.The expressions of QPRT and SHMT2 are positively correlated.
