目的:探讨C6胶质瘤大鼠模型中脑温度的空间分布特征及其在界定肿瘤浸润边界中的价值.方法:将16只SD大鼠随机分为实验组(n=8)与对照组(n=8).实验组于右侧尾状核立体定向注射C6胶质瘤细胞,对照组注射等量培养基.术后第14天,于7T磁共振成像扫描仪上行常规磁共振成像及质子磁共振波谱测温扫描,分别在肿瘤实体部、瘤周区及对侧正常脑区(尾状核)(对照组为双侧对称位置)采集波谱.通过体外温度校正模型计算各感兴趣体积的绝对脑温.结果:在胶质瘤大鼠中,不同脑区脑温差异具有统计学意义(F=37.05,P<0.01).肿瘤实体部分温度最高(38.53±0.24)℃,高于瘤周区[(38.14±0.18)℃,P<0.01]及对侧正常脑区[(37.63±0.19)℃,P<0.01];瘤周区温度亦高于对侧正常脑区(P<0.01).对照组双侧脑区温度无差异(P>0.05).实验组对侧正常脑区温度高于对照组同侧脑区(P<0.01).结论:质子磁共振波谱测温技术可敏感探测C6胶质瘤大鼠模型的脑温度梯度,瘤周区温度升高为识别肿瘤浸润边界提供了潜在标志物.

Objective:To investigate the spatial distribution characteristics of brain temperature in a rat C6 glioma model and its value in defining the tumor infiltration boundary.Methods:Sixteen Sprague-Dawley(SD)rats were randomly divided into a glioma group(n=8)and a control group(n=8).The glioma group underwent stereotactic injection of C6 glioma cells into the right caudate nucleus,while the control group received an equal volume of culture medium.On the 14th day post-injection,all rats underwent conventional MRI and proton magnetic resonance spectroscopy(1H-MRS)on a 7T MRI scanner.Spectra were acquired from the tumor solid part,peritumoral area,and contralateral normal caudate nucleus(symmetric bilateral locations for the control group).Absolute brain temperature for each volume of interest(VOI)was calculated using an in vitro temperature calibration model.Results:In glioma-bearing rats,brain temperature differed significantly among the various brain regions(F=37.05,P<0.01).The temperature was highest in the tumor solid part(38.53±0.24℃),which was significantly higher than that in the peritumoral area(38.14±0.18℃,P<0.01)and the contralateral normal brain area(37.63±0.19℃,P<0.01).The temperature in the peritumoral area was also significantly higher than that in the contralateral normal brain area(P<0.01).In the control group,there was no significant temperature difference between the bilateral brain regions(P>0.05).The temperature in the contralateral normal brain area of the glioma group was significantly higher than that in the corresponding brain area of the control group(P<0.01).Conclusion:MRS thermometry can sensitively detect brain temperature gradients in the C6 glioma model.The elevated temperature in the peritumoral area provides a potential biomarker for identifying the tumor infiltration boundary.
