甘草酸对湖羊生长性能、抗氧化功能、免疫力和肠道健康的影响OA
Effects of glycyrrhizic acid on growth performance,antioxidant function,immunity,and intestinal health in Hu sheep
试验旨在探究甘草酸的抑菌作用及其对湖羊生长性能、抗氧化能力、免疫功能及肠道健康的影响.采用打孔法测定甘草酸对致病性大肠杆菌、金黄色葡萄球菌及鼠伤寒沙门氏菌生长的影响.选取60只体重为(15.95±0.45)kg的健康湖羊,随机分为4组,每组3个重复,每个重复5只.对照组饲喂基础饲粮、试验Ⅰ组、Ⅱ组和Ⅲ组分别在基础饲粮中添加2、4和8 g/kg甘草酸.预试期7 d,正式试验期60 d.结果显示,甘草酸能够有效抑制致病性大肠杆菌和金黄色葡萄球菌的生长.与对照组相比,试验Ⅱ组和试验Ⅲ组末重显著提高(P<0.05).试验Ⅱ组血清中总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活性显著提高(P<0.05),丙二醛(MDA)含量显著降低(P<0.05);试验Ⅱ组和试验Ⅲ血清中谷胱甘肽过氧化物酶(GSH-Px)活性显著提高(P<0.05).试验Ⅱ组和试验Ⅲ组血清中白细胞介素-4(IL-4)、白细胞介素-10(IL-10)、免疫球蛋白G(IgG)、免疫球蛋白A(IgA)及回肠黏液中分泌型免疫球蛋白A(SIgA)含量显著提高(P<0.05),白细胞介素-1β(IL-1β)和白细胞介素-6(IL-6)含量显著降低(P<0.05).试验Ⅱ组和试验Ⅲ组结肠中闭锁蛋白(Occludin)、闭锁小带蛋白-1(ZO-1)、α连环蛋白(α-catenin)、黏蛋白2(Muc2)mRNA相对表达量显著提高(P<0.05),试验Ⅲ组ZO-1和Muc2的mRNA相对表达量显著高于试验Ⅱ组(P<0.05).试验Ⅱ组和试验Ⅲ组粪便中大肠杆菌含量显著降低(P<0.05).研究表明,饲粮中添加4 g/kg和8 g/kg甘草酸能够促进湖羊生长、增强抗氧化能力和免疫功能、提高肠道紧密连接蛋白表达并降低了粪便中大肠杆菌的数量.
This study aimed to investigate the antibacterial effect of glycyrrhizic acid and its effects on the growth performance,antioxidant capacity,immune function,and intestinal health of Hu sheep.The punch method was used to determine the inhibitory effect of glycyrrhizic acid on the growth of pathogenic Escherichia coli,Staphylococcus aureus,and Salmonella typhimurium.A total of 60 healthy Hu sheep with an initial body weight of(15.95±0.45)kg were randomly divided into four groups,with three replicates per group and five sheep per replicate.The control group was fed a basal diet,while experimental groups Ⅰ,Ⅱ,and Ⅲ received the basal diet supplemented with 2,4,and 8 g/kg glycyrrhizic acid,respectively.The pre-test period was seven days,and the formal test period was 60 days.The results showed that glycyrrhizic acid could effectively inhibit the growth of pathogenic Escherichia coli and Staphylococcus aureus.Compared with the control group,the final body weight of experimental groups Ⅱ and Ⅲ was significantly increased(P<0.05).In experimental group Ⅱ,serum total antioxidant capacity(T-AOC)and superoxide dismutase(SOD)activity were significantly increased(P<0.05),while malondialdehyde(MDA)content was significantly decreased(P<0.05).The glutathione peroxidase(GSH-Px)activity in serum was significantly increased in experimental groups Ⅱ and Ⅲ(P<0.05).The serum levels of interleukin-4(IL-4),interleukin-10(IL-10),immunoglobulin G(IgG),immunoglobulin A(IgA),and the ileal mucus secretory immunoglobulin A(SIgA)were significantly elevated in experimental groups Ⅱ and Ⅲ(P<0.05),while interleukin-1β(IL-1β)and interleukin-6(IL-6)levels were significantly reduced(P<0.05).The mRNA relative expression levels of Occludin,zonula occludens-1(ZO-1),α-catenin,and mucin 2(Muc2)in the colon were significantly increased in experimental groups Ⅱand Ⅲ(P<0.05).The mRNA relative expression levels of ZO-1 and Muc2 in experimental group Ⅲ were significantly higher than in experimental group Ⅱ(P<0.05).The fecal Escherichia coli count in experimental groups Ⅱ and Ⅲ was significantly lower than in the control group(P<0.05).The study shows that dietary supplementation with 4 g/kg and 8 g/kg glycyrrhizic acid can promote growth,enhance antioxidant capacity and immune function,upregulate the expression of intestinal tight junction proteins,and reduce fecal Escherichia coli counts in Hu sheep.
