牛病毒性腹泻病毒E2基因在大豆叶片中的表达及对小鼠免疫指标的影响OA
Expression of bovine viral diarrhea virus E2 gene in soybean leaves and its effects on immune indices in mice
试验旨在探究牛病毒性腹泻病毒(BVDV)E2基因在大豆叶片中的表达及其产物的免疫原性.首先,对BVDV吉林分离株E2基因的中和抗原表位序列进行分析,设计并合成多表位聚合基因序列BVDV-JZ;进而利用同源重组技术,成功构建了重组病毒SMV3-2-BVDV-JZ.逆转录聚合酶链反应(RT-PCR)、酶联免疫吸附试验(ELISA)及蛋白质印迹法(Western-blot)检测证实,BVDV-JZ多表位聚合蛋白可在转基因大豆叶片中成功表达,且该蛋白能够被BVDV阳性血清特异性识别.为评估其口服免疫效果,使用10 mg含该蛋白的大豆叶片总蛋白灌服免疫BALB/c小鼠.结果显示,BVDV-JZ多表位聚合蛋白可以在大豆叶片中表达.Western-blot检测显示,大豆叶片中表达的BVDV-JZ多表位聚合蛋白可以被BVDV阳性血清特异性识别.BALB/c小鼠灌服免疫后,未对生长产生影响,其血清中免疫球蛋白G(IgG)和粪便中免疫球蛋白A(IgA)含量显著高于空白组(P<0.05);灌服21 d后,血清中干扰素-γ(IFN-γ)和白细胞介素-4(IL-4)含量显著高于空白组(P<0.05).口服免疫后小鼠淋巴细胞增殖能力增强.研究表明,大豆可以作为生物反应器生产外源蛋白,并能诱导机体产生免疫应答,此结果为可饲疫苗的研发与应用提供参考.
This study aimed to investigate the expression of the bovine viral diarrhea virus(BVDV)E2 gene in soybean leaves and the immunogenicity of its expressed product.First,the neutralizing epitope sequence of the E2 gene from a BVDV Jilin isolate was analyzed,and a multi-epitope polymer gene sequence named BVDV-JZ was designed and synthesized.Subsequently,the recombinant virus SMV3-2-BVDV-JZ was successfully constructed using homologous recombination technology.Reverse transcription-polymerase chain reaction(RT-PCR),enzyme-linked immunosorbent assay(ELISA),and Western blot analysis confirmed that the BVDV-JZ multi-epitope protein was successfully expressed in transgenic soybean leaves and could be specifically recognized by BVDV-positive serum.To evaluate its oral immunization effect,BALB/c mice were immunized by gavage with 10 mg of total protein extracted from soybean leaves containing the target protein.The results showed that the BVDV-JZ multi-epitope protein could be expressed in soybean leaves.Western blot detection indicated that the BVDV-JZ protein expressed in soybean leaves was specifically recognized by BVDV-positive serum.After oral immunization in BALB/c mice,no adverse effects on growth were observed.The levels of immunoglobulin G(IgG)in serum and immunoglobulin A(IgA)in feces were significantly higher than those in the blank group(P<0.05).After 21 days of gavage,the serum levels of interferon-γ(IFN-γ)and interleukin-4(IL-4)were significantly higher than those in the blank group(P<0.05).Moreover,lymphocyte proliferation ability was enhanced after oral immunization.This study shows that soybean can serve as a bioreactor for producing exogenous proteins and can induce immune responses in vivo,providing a reference for the development and application of edible vaccines.
