迷迭香水提液及迷迭香酸对大肠杆菌感染小鼠保护作用研究OA
Study on protective effects of rosemary aqueous extract and rosmarinic acid on mice infected with Escherichia coli
试验旨在探究迷迭香水提液及其活性成分迷迭香酸对大肠杆菌的体外抑菌活性,以及对感染大肠杆菌小鼠肠道损伤的保护作用.采用热水回流法制备迷迭香水提液,通过牛津杯法及微量肉汤稀释法评价体外抑菌活性,并测定最小抑菌浓度(MIC);建立小鼠大肠杆菌感染模型,观察药物预处理对感染小鼠存活率、体重、肠道功能、脏器病理及血清生化指标的影响.结果显示,迷迭香水提液中迷迭香酸含量为6.73 mg/g;1.00 g/mL和0.50 g/mL迷迭香水提液对大肠杆菌为中敏,0.25 g/mL迷迭香水提液及1.0~4.0 g/L迷迭香酸为低敏;迷迭香水提液与迷迭香酸的MIC分别为0.250 00 g/mL和4.000 00 g/L;迷迭香水提液在12 h内呈现剂量依赖性的抑菌作用.体内试验表明,迷迭香水提液(0.25 g/kg和0.50 g/kg)及迷迭香酸(3.365 mg/kg)预处理可以提高感染小鼠存活率,缓解体重下降;0.50 g/kg迷迭香水提液能改善肠道蠕动亢进、减轻脏器损伤及肠道组织病理改变,并降低血清炎症因子含量与转氨酶活性.研究表明,迷迭香水提液与迷迭香酸具有抑制大肠杆菌及缓解感染所致肠道损伤的作用,具备作为畜禽大肠杆菌感染绿色防控饲料添加剂的开发潜力.
This study aimed to investigate the in vitro antibacterial activity of rosemary aqueous extract and its active component,rosmarinic acid,against Escherichia coli,as well as their protective effects on intestinal damage in infected mice.Rosemary aqueous extract was prepared using the hot water reflux extraction method.The in vitro antibacterial activity was evaluated using the Oxford cup assay and the micro-broth dilution method,and the minimum inhibitory concentration(MIC)was determined.A mouse model of Escherichia coli infection was established to observe the effects of pretreatment with the rosemary aqueous extract or rosmarinic acid on the survival rate,body weight,intestinal function,organ pathology,and serum biochemical indices of infected mice.The results showed that the rosmarinic acid content in the rosemary aqueous extract was 6.73 mg/g.Rosemary aqueous extract at 1.00 g/mL and 0.50 g/mL exhibited moderate sensitivity against Escherichia coli,while 0.25 g/mL rosemary aqueous extract and 1.0~4.0 g/L rosmarinic acid showed low sensitivity.The MIC of rosemary aqueous extract and rosmarinic acid were 0.250 00 g/mL and 4.000 00 g/L respectively.The rosemary aqueous extract demonstrated dose-dependent antibacterial activity within 12 hours.In vivo experiments revealed that pretreatment with rosemary aqueous extract(0.25 g/kg and 0.50 g/kg)or rosmarinic acid(3.365 mg/kg)improved the survival rate and alleviated weight loss in infected mice.The 0.50 g/kg rosemary aqueous extract ameliorated intestinal hypermotility,reduced organ damage and intestinal histopathological alterations,and reduced serum inflammatory factor content and transaminase activity.The study shows that rosemary aqueous extract and rosmarinic acid exhibit inhibitory effects against Escherichia coli and mitigate infection-induced intestinal damage,demonstrating potential as green feed additives for the prevention and control of Escherichia coli infections in livestock and poultry.
