少腹逐瘀丸对子宫内膜异位症大鼠MMP-9及TIMP-2表达的影响OA
Effects of Shaofu Zhuyu Pills on MMP-9 and TIMP-2 expression in rats with endometriosis
目的 分析少腹逐瘀丸对子宫内膜异位症(EMs)大鼠基质金属蛋白酶-9(MMP-9)及基质金属蛋白酶组织抑制因子-2(TIMP-2)表达的影响.方法 将雌性大鼠随机均分为假手术组、模型组、低剂量组(2.8 g/kg少腹逐瘀丸)、高剂量组(5.6 g/kg少腹逐瘀丸)和孕三烯酮组(1.25 mg/kg孕三烯酮).除假手术组外,其余组均建立EMs模型.比较各组血清肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β、IL-6、雌二醇(E2)、孕激素(P)、促卵泡激素(FSH)、促黄体生成素(LH)水平.采用HE染色观察病理变化,比较各组子宫内膜组织MMP-9、TIMP-2 mRNA和蛋白表达水平.结果 与假手术组比较,EMs组大鼠子宫内膜组织出现明显病理损伤,而经少腹逐瘀丸干预后子宫内膜组织病理损伤明显改善(P<0.05).高剂量组和孕三烯酮组异位病灶质量、体积低于模型组(P<0.05).与假手术组比较,模型组、低剂量组、高剂量组和孕三烯酮组血清TNF-α、IL-6、IL-1β、E2、P以及MMP-9 mRNA和蛋白表达水平升高,FSH、LH以及TIMP-2 mRNA和蛋白表达水平降低(P<0.05);与模型组比较,高剂量组和孕三烯酮组上述趋势得到逆转(P<0.05).结论 少腹逐瘀丸干预EMs大鼠可明显改善子宫内膜组织病理损伤,减轻炎症反应,改善性激素水平,其作用可能通过下调MMP-9表达、上调TIMP-2表达实现.
Aim To analyze the effects of Shaofu Zhuyu Pills on the expression of matrix metalloproteinase-9(MMP-9)and tissue inhibitor of metalloproteinase-2(TIMP-2)in rats with endometriosis(EMs).Methods Female rats were randomly divided into a sham-operated group,a model group,a low-dose group(2.8 g/kg Shaofu Zhuyu Pills),a high-dose group(5.6 g/kg Shaofu Zhuyu Pills),and a gestrinone group(1.25 mg/kg gestrinone).EMs models were established in all groups except the sham-operated group.Serum levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),IL-6,estradiol(E2),progesterone(P),follicle-stimulating hormone(FSH),and luteinizing hormone(LH)were compared.Pathological changes were observed by HE staining,and the mRNA and protein expression levels of MMP-9 and TIMP-2 in endometrial tissue were compared.Results Compared with the sham-operated group,the EMs model group showed significant pathological damage in the endometrial tissue.However,intervention with Shaofu Zhuyu Pills significantly improved this pathological damage(P<0.05).The mass and volume of ectopic lesions in the high-dose group and the gestrinone group were lower than those in the model group(P<0.05).Compared with the sham-operated group,the model,low-dose,high-dose,and gestrinone groups showed increased serum levels of TNF-α,IL-6,IL-1β,E2,P,and elevated mRNA and protein expression of MMP-9,while levels of FSH,LH,and mRNA and protein expression of TIMP-2 were decreased(P<0.05).Compared with the model group,these trends were reversed in the high-dose and gestrinone groups(P<0.05).Conclusion Shaofu Zhuyu Pill intervention in EMs rats can significantly improve pathological damage in en-dometrial tissue,alleviate inflammatory responses,and normalize sex hormone levels.Its mechanism of action may involve the down-regulation of MMP-9 expression and upregulation of TIMP-2 expression.
