目的 探究乳酸(Lac)减轻大鼠创伤性脑损伤(TBI)的作用机制.方法 将SPF级雄性SD大鼠 50 只随机均分为假手术(Sham)组、TBI 组、Lac 组、核苷酸结合结构域富含亮氨酸重复序列和含热蛋白结构域受体 3(NLRP3)抑制剂(MCC950)组、Lac+MCC950 组.比较各组大鼠改良神经功能缺损评分(mNSS)、脑组织病理变化、神经细胞凋亡率和脑组织神经元核蛋白(NeuN)与NLRP3 共表达、炎症因子水平以及NLRP3/半胱氨酸天冬氨酸蛋白酶-1(caspase-1)信号通路相关蛋白的表达水平.结果 与Sham组比较,TBI组脑组织病理损伤显著,炎症细胞浸润明显,NeuN与NLRP3 共表达荧光强度增强,mNSS、细胞凋亡率、炎症因子和 NLRP3/caspase-1 信号通路相关蛋白表达水平均升高(P<0.05).与 TBI 组比较,Lac 组、MCC950 组、Lac+MCC950 组逆转上述指标水平(P<0.05);与Lac组比较,MCC950 组、Lac+MCC950 组进一步逆转上述指标水平(P<0.05).结论 Lac能够有效减轻TBI大鼠的神经功能缺损与组织病理损伤,抑制大鼠神经细胞焦亡,其作用机制可能与抑制NLRP3/caspase-1信号通路有关.

Aim To explore the mechanism by which lactate acid(Lac)alleviates traumatic brain injury(TBI)in rats.Methods Fifty SPF grade male SD rats were randomly divided into a sham surgery(Sham)group,a TBI group,a Lac group,a nu-cleotide-binding domain leucine-rich repeat and pyrin domain-containing receptor 3(NLRP3)inhibitor(MCC950)group,and a Lac+MCC950 group.The modified Neurological Severity Score(mNSS),pathological changes in brain tissue,neuronal apoptosis rate,as well as the co-expression of neuronal nucleir protein(NeuN)and NLRP3,levels of inflammatory factors,and expression levels of NLRP3/cysteine aspartic acid specific protease-1(caspase-1)signaling pathway related proteins in brain tissue were compared among different groups of rats.Results Compared with the Sham group,the TBI group showed significant pathological damage to brain tissue,obvious infiltration of inflammatory cells,enhanced fluorescence intensity of NeuN and NLRP3 co-expression,the mNSS,cell apoptosis rate,and the levels of inflammatory factors and NLRP3/caspase-1 signaling pathway related protein in brain tissue were all increased(P<0.05).Compared with the TBI group,the Lac group,MCC950 group,and Lac+MCC950 group reversed the changes of the above indicators(P<0.05);Compared with the Lac group,the MCC950 group and the Lac+MCC950 group further reversed the alterations of the above indicators(P<0.05).Conclusion Lac can effectively alleviate neurological deficits and histopathological damage in rats after TBI,inhibit neuronal apoptosis,and its mechanism of action may be related to the inhibition of the NLRP3/caspase-1 signaling pathway.
