丰富环境上调m6A修饰促进小鼠视皮质突触可塑性相关基因蛋白的表达OA
Enriched environment upregulates m6A modification and promotes the expression of synaptic plasticity-related genes in the mouse visual cortex
目的 探讨丰富环境(EE)对小鼠视皮质m6A修饰及突触可塑性相关基因蛋白表达的影响.方法 14 只8 周龄雄性C57BL/6J小鼠随机均分为对照组(CON组)和EE组.通过斑点杂交和免疫组织化学检测小鼠视皮质m6A修饰水平;Western blotting检测甲基转移酶和去甲基化酶;qRT-PCR检测编码突触蛋白的mRNA,Western blotting检测突触蛋白水平.结果 与CON组相比,EE组小鼠m6A修饰水平上升,m6A去甲基化酶ALKBH5 下调(P<0.05),其他甲基化修饰酶蛋白表达两组间比较差异无显著性(P>0.05).与CON组小鼠相比,EE组小鼠编码突触蛋白SYP、PSD-95 和Homer-1的mRNA水平差异无显著性(P>0.05);而SYP、PSD-95 和Homer-1的蛋白水平均上调(P<0.05).结论 丰富环境上调小鼠视皮质可塑性相关基因m6A修饰,促进其蛋白翻译,进而引起突触蛋白表达的上调.
Aim To investigate the effects of enriched environment(EE)on m6A modification and the expression of synap-tic plasticity-related genes and proteins in the mouse visual cortex.Methods Fourteen 8-week-old male C57BL/6J mice were randomly divided into a control group(CON group)and an EE group.Dot blot hybridization and immunohistochemistry were used to detect m6A modification levels in the visual cortex.Western blotting was performed to analyze the expression of methyltransferases and demethylases.qRT-PCR was used to measure mRNA levels of synaptic protein-encoding genes,and Western blotting was applied to detect synaptic protein expression.Results Compared with the CON group,the EE group showed increased m6A modification levels and downregulated expression of the m6A demethylase ALKBH5(P<0.05),while no significant differences were observed in other methylation-related enzymes(P>0.05).Although no significant differences were found in the mRNA levels of synaptic protein genes SYP,PSD-95,and Homer-1 between the two groups(P>0.05),the protein levels of SYP,PSD-95,and Homer-1 were significantly up-regulated in the EE group(P<0.05).Conclusion Enriched environment upregulates m6A modification of plasticity-related genes in the mouse visual cortex,thereby promoting their protein translation and leading to increased expression of synaptic proteins.
