Role and molecular mechanisms of CMTM4 in colorectal carcinogenesis
目的 探讨人类趋化素样因子家族(chemokine-like factor family member,CMTM)在结直肠癌(colorectal cancer,CRC)发生中的作用及其分子机制.方法 采用免疫组化和RT-PCR检测CMTM4在CRC和正常组织中的表达,评估CMTM4表达与患者临床病理特征及预后的关系.通过CCK-8细胞增殖实验和Transwell迁移与侵袭实验,评估CMTM4对CRC细胞增殖、迁移及侵袭能力的影响.结果 免疫组化和RT-PCR结果表明,CMTM4在CRC组织中的表达显著低于正常组织,CMTM4低表达与CRC的T分期、Duke分期及不良预后相关.CCK-8实验显示,CMTM4过表达显著抑制CRC细胞增殖;Transwell实验结果显示,CMTM4过表达显著降低细胞的迁移和侵袭能力.对CMTM4共表达基因进行GO/KEGG分析,结果显示CMTM4参与免疫相关和代谢相关途径,CMTM4表达与免疫细胞浸润水平和多种免疫细胞标志物集的表达呈正相关.结论 CMTM4参与结直肠癌的发生、发展,可能作为CRC的潜在分子治疗靶点.
Objective To investigate the role of chemokine-like factor family member 4(CMTM4)in the devel-opment of colorectal cancer(CRC)and to explore its underlying molecular mechanisms.Methods The expression of CMTM4 in CRC tissues and normal colorectal tissues was examined using immunohistochemistry and reverse transcrip-tion polymerase chain reaction(RT-PCR).The associations between CMTM4 expression and clinicopathological pa-rameters as well as patient prognosis were analyzed.The effects of CMTM4 on CRC cell proliferation,migration,and invasion were evaluated CCK-8 proliferation assays and Transwell migration and invasion assays.In addition,Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analyses were performed on CMTM4 co-expressed genes,and correlations between CMTM4 expression and immune cell infiltration were assessed.Results Immunohistochemistry and RT-PCR analyses demonstrated that CMTM4 expression was significantly lower in CRC tis-sues than in normal tissues.Low CMTM4 expression was significantly associated with advanced T stage,Duke stage,and poor prognosis in patients with CRC.CCK-8 assays showed that CMTM4 overexpression significantly inhibited CRC cell proliferation.Transwell assays revealed that CMTM4 overexpression markedly reduced the migratory and in-vasive abilities of CRC cells.GO and KEGG analyses indicated that CMTM4 was mainly involved in immune-related and metabolism-related pathways.Furthermore,CMTM4 expression was positively correlated with immune cell infiltra-tion levels and with the expression of multiple immune cell marker gene sets.Conclusion CMTM4 is involved in the development and progression of colorectal cancer and may serve as a potential molecular therapeutic target for CRC.
