Expression of PAIP1 in colorectal adenocarcinoma and its role in cell proliferation
目的 探讨PAIP1表达与结肠腺癌临床病理特征与预后的关系及其在结肠腺癌细胞增殖中的作用.方法 采用免疫组化SP法检测83例结肠腺癌和癌旁非瘤组织中PAIP1蛋白的表达,分析PAIP1蛋白表达与结肠腺癌临床病理特征的关系.应用Kaplan-Meier生存法分析PAIP1蛋白表达与结肠腺癌患者生存期之间的相关性;运用Cox回归模型评估PAIP1在结肠腺癌预后风险评估中的作用.转染PAIP1沉默质粒(shRNA),利用Western blot实验验证沉默效率.通过CCK-8、EdU和克隆形成实验检测PAIP1沉默对结肠腺癌细胞增殖能力的影响.结果 免疫组化显示PAIP1蛋白在结肠腺癌组织中的阳性率和强阳性率分别为92.78%(77/83)和48.19%(40/83),显著高于癌旁非瘤组织(4.82%、1.20%).χ2检验显示PAIP1蛋白表达与结肠腺癌患者病理分级密切相关(P=0.011).Kaplan-Meier生存分析表明,PAIP1蛋白高表达的结肠腺癌患者总生存期低于PAIP1蛋白低表达患者(P=0.003 7).Cox多因素回归分析表明,淋巴结转移(HR=2.918,95%CI=1.405～6.060,P=0.004)和PAIP1蛋白强阳性(HR=2.142,95%CI=1.037～4.427,P=0.040)是影响结肠腺癌患者预后的独立风险因素.CCK-8、EdU和克隆形成实验显示PAIP1沉默可抑制结肠腺癌细胞增殖能力.结论 PAIP1在结肠腺癌中高表达,且与患者不良预后密切相关;沉默PAIP1可抑制结肠腺癌细胞增殖能力.
Objective To evaluate the expression of PAIP1 in colorectal adenocarcinoma,analyze its correlation with clinicopathological features and prognosis,and identify the role of PAIP1 in the proliferation of colorectal adeno-carcinoma cells.Methods PAIP1 protein expression was detected in 83 cases of colorectal adenocarcinoma tissue and paired adjacent non-tumor tissue using the immunohistochemical SP method.Then,the relationship between PAIP1 protein expression and clinicopathological features of colorectal adenocarcinoma was analyzed.The Kaplan-Meier method was employed to analyze the correlation between PAIP1 protein expression and the survival of colorectal adenocarcinoma patients.The Cox regression model was used to assess the role of PAIP1 in predicting the risk of colorectal adenocarcinoma.Lentivirus-mediated shRNA was used to silence PAIP1 in colorectal adenocarcinoma cells.Then,the effects of PAIP1 silencing on the proliferative ability of colorectal adenocarcinoma cells were deter-mined by CCK-8,EdU,and colony formation assays.Results The positive and strong positive rates of PAIP1 protein in colorectal adenocarcinoma tissues were 92.78%(77/83)and 48.19%(40/83),respectively,significantly higher than in adjacent non-tumor tissues(4.82%,1.20%).Chi-squared test results showed that PAIP1 protein expression was significantly related to the pathological grade(P=0.011).Kaplan-Meier survival analysis indicated that patients with high expression of PAIP1 protein had a significantly lower overall survival than those with low expression(P=0.003 7).Multivariate Cox regression analysis showed that lymph node metastasis(HR=2.918,95%CI=1.405-6.060,P=0.004)and strong positive expression of PAIP1(HR=2.142,95%CI=1.037-4.427,P=0.040)were inde-pendent risk factors for colorectal adenocarcinoma.CCK-8,EdU,and colony formation assay showed that knockdown of PAIP1 inhibited the proliferation capacity of colorectal adenocarcinoma cells.Conclusion PAIP1 is highly ex-pressed in colorectal adenocarcinoma and is closely related to poor prognosis.PAIP1 affects cell proliferation of colorectal adenocarcinoma.
