犹素化修饰在肿瘤发生发展中作用的研究进展OA

Research advance on UFMylation in tumorigenesis and progression

犹素化修饰(UFMylation)是一种新型类泛素化修饰,由特异性的活化酶E1(UBA5)、结合酶E2(UFC1)和连接酶E3(UFL1)级联催化成熟的泛素折叠修饰因子(ubiquitin-fold modifier 1,UFM1)共价连接到底物蛋白,调控靶蛋白功能.研究表明犹素化修饰在调控蛋白质的稳定性、内质网应激和DNA损伤修复等生物学过程中发挥着重要作用,其调控异常与人类多种肿瘤的发生发展密切相关.犹素化修饰一方面能通过激活促癌信号通路来加速肿瘤的发生发展,另一方面可以通过维持基因组稳定性和抑制癌基因活性发挥抑癌作用,说明犹素化修饰在肿瘤调控中发挥双重作用.文章综述了犹素化修饰的发生过程、生物学功能及其在肿瘤发生发展中的作用,探讨了犹素化修饰在肿瘤靶向治疗中的应用前景.

UFMylation is a novel ubiquitin-like modification.In this process,the mature ubiquitin-fold modifier 1(UFM1)molecule is covalently conjugated to substrate proteins via a cascade catalyzed by the specific activating enzyme E1(UBA5),conjugating enzyme E2(UFC1),and ligase E3(UFL1),thereby regulating the functions of target proteins.Research indicates that UFMylation plays significant roles in various biological processes,including the regulation of protein stability,endoplasmic reticulum stress,and DNA damage repair.Abnormal regulation of UFMylation is closely associated with the development of various human tumors.On one hand,UFMylation can accelerate tumor progression by activating oncogene signaling pathways,on the other hand,it can exert tumor-suppressive effects by maintaining genomics stability and inhibiting oncogene activity,suggesting a dual role of UFMylation in tumor regulation.This paper reviews the mechanism,biological functions of UFMylation,and its role in tumor development,while also discussing the prospects of targeting UFMylation for cancer therapy.
