Influence of Genotype on the Anther Culture in Capsicum annuum L.

研究以48份不同基因型辣椒栽培种为材料开展花药胚状体诱导及组培苗再生试验,以期筛选高花药培养力辣椒材料,并观察其胚状体的形态发生过程.结果表明:48 份不同基因型辣椒材料的花药膨大率、花药褐化率、胚状体诱导率和组培苗再生率变异系数分别为 70.63%、28.60%、101.27%、87.86%;其中,花药膨大率与褐化率显著正相关,与胚状体诱导率、组培苗再生率极显著正相关;花药褐化率分别与胚状体诱导率、组培苗再生率极显著正相关,胚状体诱导率与组培苗再生率极显著正相关.试验筛选到 1 份高花药培养力基因型 22Y19,其胚状体诱导率达 20.14%,组培苗再生率达 42.25%,其胚状体发育经历了球形胚、心形胚、鱼雷形胚和子叶形胚 4 个典型阶段,并最终发育成完整的植株.该材料不仅可作为桥梁亲本用于提升低诱导率基因型的花药培养效率,也可作为关键试验材料用于深入解析胚状体发育的分子调控机制.

This study utilized 48 distinct genotypes of Capsicum annuum L.cultivars for anther embryoid induction and regenerated plantlet culture test,aiming to screen the genotypes with superior androgenic competence and unveil the morphogenetic process of embryoids.The results revealed that the coefficients of variation of anther swelling rate,browning rate,embryoid induction rate,and plantlet regeneration rate reached 70.63%,28.60%,101.27%,and 87.86%,respectively,among the 48 genotypes.The anther swelling rate had a significantly positive correlation with browning rate and extremely significantly positive correlations with embryoid induction rate and plantlet regeneration rate.The browning rate showed extremely significant positive correlations with embryoid induction rate and plantlet regeneration rate.Furthermore,the embryoid induction rate and plantlet regeneration rate exhibited an extremely significantly positive correlation.The screening process identified genotype 22Y19 as an outstanding androgenic performer,achieving an embryoid induction rate of 20.14%and a plantlet regeneration rate of 42.25%.Developmental tracking revealed that embryogenic progression of 22Y19 through four distinct morphological stages:globular,heart-shaped,torpedo,and cotyledonary embryos,culminating in complete plant formation.This genotype could not only be used as a bridge parent to enhance the efficiency of anther culture for genotypes with low induction rates but also as a key experimental material for deciphering the molecular mechanism of embryoid development.
