Genetic Diversity Analysis of Aromatic Rice Germplasm Resources from South China Based on SSR and ISSR Markers

采用SSR和ISSR分子标记技术,对 20 份华南地区香稻材料的遗传多样性进行了分析,揭示不同材料间的遗传特征.结果表明:SSR标记体系筛选的 8 对引物共扩增出 63 条条带,其中 41条条带表现多态性,多态条带比率平均为 63.07%,材料的遗传相似系数为 0.382 9~0.969 6,平均值为 0.676 3,呈中等多态性.ISSR标记体系筛选 5 条引物扩增出 38 条带,其中15条带具有多态性,多态条带比率平均为41.80%,材料间遗传相似系数为0.714 2~0.985 9,平均值达0.850 1,多态性较低.SSR标记检测到的多态性显著高于ISSR,更适合香稻遗传多样性分析.将 2 种分子标记得出的数据进行聚类分析,均将供试材料分为 3 大类群:类群Ⅰ包含 17 份材料,类群Ⅱ和Ⅲ分别包含 1 份和 2 份材料,2 种标记在主要类群划分上具有一致性,但在部分材料(象香丝苗、美香占)的归类上存在差异,表明栽培稻遗传基础狭窄,且材料之间遗传存在一定差异.

This study systematically analyzed the genetic diversity of 20 aromatic rice germplasm accessions from South China based on SSR and ISSR markers.The results showed that the amplification with eight pairs of SSR primers produced 63 bands,among which 41 bands showed polymorphism,with an average polymorphism rate of 63.07%.The genetic similarity coefficients ranged from 0.382 9 to 0.969 6,with a mean value of 0.676 3,indicating moderate polymorphism.Five ISSR primers produced 38 bands,among which 15 bands exhibited polymorphism,with an average polymorphism rate of 41.80%.The genetic similarity coefficients ranged from 0.714 2 to 0.985 9,with a mean value of 0.850 1,indicating low polymorphism.SSR markers detected higher polymorphism than ISSR markers,being more suitable for the genetic diversity analysis of aromatic rice.Cluster analysis classified the accessions into three major groups.Group I contained 17 accessions,while Groups II and III consisted of 1 and 2 accessions,respectively.SSR and ISSR markers were consistent in the classification of most accessions but had differences in the classification of some accessions(such as'Xiangxiangsimiao'and'Meixiangzhan'),which indicated that the genetic basis of cultivated rice was narrow and there were genetic differences among accessions.
