基于UPLC-MS/MS代谢组学技术探究乳杆菌对神经递质合成的影响OA
Effect of Lactobacillus on neurotransmitter synthesis based on UPLC-MS/MS metabolomics technology
目的:探究两种不同乳杆菌对外周神经递质的影响.方法:选取8周龄C57BL/6雄性小鼠18只适应性喂养1周后,随机分为对照(DZ)组、罗伊氏乳杆菌(LYS)组、嗜酸乳杆菌(SS)组,每组6只.DZ组每天灌胃生理盐水0.2 mL/只,干预组(罗伊氏乳杆菌、嗜酸乳杆菌组)每天分别灌胃0.2 mL/只的对应菌种冻干粉溶液.灌胃10周后采集小鼠血液,采用UPLC-MS/MS对小鼠血清中的神经递质进行定性定量分析.同时通过多元统计分析观察各组小鼠血清中神经递质含量的变化,并结合t检验(P<0.05)与OPLS-DA(VIP>1)筛选出差异代谢物.结果:小鼠血清中检测到5类共39种神经递质,其中氨基酸类21种,单胺类13种,胆碱类1种,其他类4种.多元统计分析显示9种差异代谢物.通过罗伊氏乳杆菌和嗜酸乳杆菌干预后ILA(P<0.01)、IAA(P<0.01)含量均明显上升,GABA(P<0.05)含量明显下降.同时,在不同乳杆菌干预后发现SS组中IAM(P<0.05)、5-HIAA(P<0.05)、Pro(P<0.01)含量明显升高,Trp(P<0.01)、Gln(P<0.000 1)含量明显降低,而LYS组中XA(P<0.001)含量明显升高.结论:两种乳杆菌均可促进兴奋性神经递质释放,减少抑制性神经递质积累,但两种乳杆菌对其他神经递质的积累存在一定的差异,研究结果可为乳杆菌改善疾病的相关研究提供理论与实验依据.
Objective:To investigate the effects of 2 different Lactobacillus species on peripheral neurotransmitters.Methods:A total of 18 8-week-old C57BL/6 male mice were selected for adaptive feeding for 1 week and randomly divided into the control(DZ),group the Lactobacillus reuteri(LYS)group,and the Lactobacillus acidophilus(SS)group,with 6 mice in each group.The DZ group was gavaged with 0.2 mL/each of saline per day,and the intervention groups(Lactobacillus reuteri and Lactobacil-lus acidophilus groups)were gavaged with 0.2 mL/each of lyophilized powdered solution of the corresponding strains per day,re-spectively.The blood of mice was collected after 10 weeks of gavage,and the neurotransmitters in mouse serum were analyzed qualitatively and quantitatively by UPLC-MS/MS.Meanwhile,the changes of neurotransmitter contents in the serum of mice in each group were observed by multivariate statistical analysis,and the differential metabolites were screened out by combining t-test(P<0.05)with OPLS-DA(VIP>1).Results:A total of 39 neurotransmitters from 5 classes were detected in mouse serum,in-cluding 21 amino acids,13 monoamines,1 choline,and 4 others.Multivariate statistical analysis revealed 9 differential metabo-lites.The content of ILA(P<0.01)and IAA(P<0.01)was significantly increased,and the content of GABA(P<0.05)was significantly decreased after the intervention by Lactobacillus reuteri and Lactobacillus acidophilus.Meanwhile,IAM(P<0.05),5-HIAA(P<0.05),and Pro(P<0.01)contents were found to be significantly higher,and Trp(P<0.01)and Gln(P<0.000 1)contents were significantly lower in the SS group after different Lactobacillus interventions,whereas XA(P<0.001)contents were found to be significantly higher in the LYS group.Conclusion:Both species of Lactobacillus can promote the release of excit-atory neurotransmitters and reduce the accumulation of inhibitory neurotransmitters,but there are some differences between the two species of Lactobacillus in the accumulation of other neurotransmitters,and the results of the study can provide theoretical and experimental bases for studies related to Lactobacillus to improve the disease.
