Elucidation of the Toxicological Mechanism of Herbicides in Morchella sextelata Based on Transcriptomic Analysis

以'六妹'羊肚菌(Morchella sextelata)M6611菌株为试材,采用PDA平板培养法并辅以RNA-seq转录组分析,研究了 4种常用除草剂(氟乐灵、二甲戊灵、精异丙甲草胺和乙草胺)对其菌丝生长的影响,以期为分析除草剂对羊肚菌的毒理机制提供参考依据.结果表明:所有除草剂均抑制菌丝生长,其中精异丙甲草胺抑制程度相对较轻;转录组分析显示除草剂处理显著干扰糖酵解/糖异生及乙醛酸循环等与能量代谢密切相关的通路.因此,在羊肚菌栽培中应谨慎使用除草剂,并加强土壤除草剂残留监测,以减少菌丝生长受阻和产量下降的潜在风险.

Taking the Morchella sextelata strain M6611 as the test material,the PDA plate culture method was adopted and combined with RNA-seq transcriptome analysis to study the effects of four commonly used herbicides(trifluralin,pendimethalin,S-metolachlor and acetochlor))on the mycelial growth of the mushroom,in order to provide references for analyzing the toxicological mechanism of herbicides on morel mushrooms.The results showed that all herbicides suppressed mycelial growth,with S-metolachlor exhibiting relatively weaker inhibition.Comprehensive transcriptome analysis revealed that herbicide treatment severely impaired critical energy metabolism pathways,particularly the processes of glycolysis/gluconeogenesis and the glyoxylate cycle.In conclusion,herbicides should be used cautiously in M.sextelata cultivation,and soil herbicide residue monitoring should be enhanced to reduce potential risks of mycelial growth inhibition and yield loss.
