基于SSR标记与形态学的白掌F1代杂种真实性鉴定OA
Authenticity Identification of Spathiphyllum kochii F1 Hybrids Using SSR Markers and Morphology
以白掌的9个亲本、8个杂种F1代以及"星光"×"美酒"的39个杂种F1代群体植株为试材,采用多态性SSR标记分析与形态学观测相结合的方法,研究了 SSR标记与形态学方法在白掌F1代杂种真实性鉴定中的效率差异,建立高效、准确的早期鉴定技术体系,以期为白掌育种提供参考依据.结果表明:从16对多态性SSR引物中筛选出8对具有父本特异性或双亲互补型条带的引物,可用于杂种鉴定.8个杂交后代的SSR扩增均显示父本或双亲基因型,鉴定真实率为100%;形态学观测仅能明确鉴定其中6个后代("青美""星辉""维美""阳萌1号""阳萌2号"和"维墨")为真杂种,"玉手"与"萌香"难以从形态区分,形态学鉴定效率为75%.SSR标记鉴定效率显著高于形态学方法,尤其适用于未表现明显父本性状的真杂种.进一步利用P14标记对"星光"×"美酒"组合的39个后代检测,均扩增出父本带型,真实率为100%,再次验证了 SSR标记的高效性与可靠性.综上,SSR标记鉴定准确、高效,可作为白掌杂交后代早期真实性鉴定的关键技术.
Taking nine parental lines of Spathiphyllum kochii,eight hybrid F1 progenies,and a population of 39 hybrid F1 plants from the cross'Xingguang'×'Meijiu'as the test materials,polymorphic SSR marker analysis with morphological observation were combined to compare the efficiency of SSR markers and morphological methods in authenticating the hybridity of Spathiphyllum kochii F1 progenies,in order to establish an efficient and accurate early identification system to support Spathiphyllum kochii breeding.The results showed that eight pairs of SSR primers with paternal specific or complementary parental bands were selected from 16 polymorphic primers for hybrid identification.SSR amplification of the eight hybrid progenies all displayed paternal or biparental genotypes,achieving an authentication rate of 100%.Morphological observation clearly identified only six of the progenies('Qingmei''Xinghui''Weimei''Yangmeng 1''Yangmeng 2'and'Weimo')as true hybrids,while'Yushou'and'Mengxiang'could not be reliably distinguished based on morphology,giving a morphological identification efficiency of 75%.The efficiency of SSR markers was significantly higher than that of morphological methods,especially for true hybrids that did not exhibit obvious paternal traits.Further detection using the P14 marker on the 39 progenies from the'Xingguang'×'Meijiu'cross all produced paternal band patterns,with a true hybrid rate of 100%,reconfirming the high efficiency and reliability of SSR markers.In conclusion,SSR marker identification is accurate and efficient,and can serve as a key technique for early authenticity assessment of Anthurium hybrid progenies.
