菜心Brassica oxyrrhina胞质不育系的转育研究OA
Development of a Cytoplasmic Male-Sterile Line in Flowering Chinese Cabbage(Brassica rapa ssp.parachinensis)through Alloplasmic Substitution with Brassica oxyrrhina Cytoplasm
以甘蓝型油菜Brassica oxyrrhina细胞质雄性不育系为试材,通过与菜心进行种间杂交和连续回交转育,结合胚拯救和快速育种技术,成功创制败育稳定的菜心Brassica oxyrrhina胞质雄性不育系,并系统研究其形态特征、花粉育性、结实特性及胞质特异性分子标记.结果表明,经6代回交,获得雄蕊退化(花丝短缩、花药干瘪)、蜜腺正常、营养和花器形态与菜心亲本一致的不育系;醋酸洋红染色显示其花粉败育,呈浅棕色、无核、形态异常;扫描电镜观察证实不育花粉外壁无孢粉素沉积、萌发沟萎缩,符合典型细胞质雄性不育特征.田间结实试验表明,不育系自交不结实,自由授粉后平均单角果结籽数达12粒,具有良好异交结实能力.基于线粒体基因组比对,成功开发特异性KASP标记KOX3,可稳定区分Brassica oxyrrhina胞质(C/C)与其他胞质类型(T/T).该不育系的获得为菜心杂种优势利用提供了关键种质资源,其配套分子标记为胞质快速鉴定与不育系纯度鉴定提供了技术支撑.
Using a cytoplasmic male sterile(CMS)line of Brassica napus carrying cytoplasm from Brassica oxyrrhina as the maternal parent,we developed a stable CMS line in flowering Chinese cabbage(Brassica rapa subsp.parachinensis)through interspecific hybridization followed by six backcrosses,assisted by embryo rescue and a speed breeding system.The line was comprehensively characterized for morphological traits,pollen fertility,seed-setting performance,and cytoplasm-specific molecular markers.The CMS line exhibited typical floral morphology of flowering Chinese cabbage,featuring degenerated stamens with shortened filaments and shriveled anthers while nectaries developed normally.Acetic acid-carmine staining revealed aborted pollen grains,characterized by small,round,anucleate,and stained pale brown.Scanning electron microscopy further showed a lack of sporopollenin deposition and the collapse of germination apertures in the sterile pollen,consistent with classical CMS phenotypes.Field evaluations confirmed complete self-sterility;however,under open pollination,plants produced an average of 12 seeds per silique,indicating strong outcrossing compatibility.Comparative mitochondrial genome analysis enabled the development of a cytoplasm-specific KASP marker,KOX3,which reliably discriminates cytoplasm from Brassica oxyrrhina(C/C genotype)from other cytoplasmic types(T/T genotype).This novel CMS line provides a valuable germplasm resource for heterosis breeding in flowering Chinese cabbage,and the KOX3 marker offers a robust molecular tool for rapid cytoplasm identification and purity assessment of CMS lines.
