Research progress on the involvement of autophagy in the pathogenesis of systemic lupus erythematosus

系统性红斑狼疮(systemic lupus erythematosus,SLE)是一种好发于育龄期女性,异质性强,致残率高,可产生多种抗体并累及多脏器的慢性自身免疫性疾病,SLE发病机制复杂,涉及遗传、环境和免疫学等诸多因素.自噬通过溶酶体降解自身受损胞质成分,控制细胞应激、蛋白质稳态和线粒体质量,其调节异常可导致细胞稳态破坏,引发多种疾病.自噬在维持固有免疫和适应性免疫稳态中发挥至关重要的作用.近年来,越来越多的研究证实,自噬参与了 SLE发病基因易感性及免疫调节紊乱的诸多环节,免疫细胞自噬异常与SLE发病密切相关.因此,深入研究自噬对SLE免疫细胞分化及功能的影响,可为寻找SLE有效治疗靶点提供新思路.

Systemic lupus erythematosus(SLE)is a chronic autoimmune disease that predominantly affects women of childbearing age.It is highly heterogeneous,characterized by a high disability rate,the pro-duction a variety of antibodies,and involvement of multiple organs.Autophagy controls cell stress,protein ho-meostasis,and mitochondrial quality by degrading damaged cytoplasmic components through lysosomes,and its dysregulation can disrupt cell homeostasis and contribute to a variety of diseases.Autophagy plays a crucial role in maintaining the homeostasis of innate and adaptive immunity.The pathogenesis of SLE is complex,in-volving genetics,environmental,and immunological factors.In recent years,an increasing number of studies have confirmed that autophagy is implicated in the genetic susceptibility to SLE and the disorder of immune reg-ulation.Therefore,in-depth research on the impact of autophagy on the differentiation of immune cells in SLE may provide new insights for identifying effective therapeutic targets for SLE.
