基于网络药理学和分子对接技术探究血满草治疗骨折的作用机制OA

Investigating the Mechanism of Action of Sambucus adnata Wall.in the Treatment of Bone Fracture Based on Network Pharmacology and Molecular Docking Technology

目的:通过网络药理学预测血满草治疗骨折的核心成分、靶点及通路,借助分子对接技术进行验证并初步探讨作用机制.方法:通过文献研究血满草的化学成分,并结合TCMSP、PubChem等数据库筛选出活性成分并得到对应靶点,再与从GeneCards等数据库中获取的骨折疾病靶点相映射获取交集靶点;利用Cytoscape 3.10.0软件和String数据库构建"中药-活性成分-疾病靶点"网络及PPI网络;利用DAVID数据库进行GO功能分析和KEGG信号通路富集分析.最后通过分子对接技术进行活性成分和关键靶点对接验证.结果:共获得血满草主要活性成分17个及其对应靶点357个,骨折疾病靶点2265个,交集靶点127个,进一步筛选得到核心活性成分5个和核心靶点12个;富集分析结果显示,血满草治疗骨折的关键通路为cAMP信号传导途径、cGMP-PKG信号通路、JAK-STAT信号通路等.分子对接结果表明,血满草的5个核心成分与关键核心靶点有较高的结合活性.结论:血满草主要通过多成分-多靶点-多通路过程治疗骨折,主要作用于MMP9、PTGS2、AKT1、ESR1、EGFR等靶点,通过调控cAMP、cGMP-PKG、JAK-STAT等通路,发挥镇痛抗炎、活血化瘀、骨细胞增殖及成骨分化等作用治疗骨折,为该药的深入研究和临床应用提供理论依据.

Objective:The core components,targets and pathways of Sambucus adnata Wall.for the treatment of bone fracture were predicted by network pharmacology,validated with the help of molecular docking technology and initially explored the mecha-nism of action.Methods:Through literature research on the chemical composition of Sambucus adnata Wall,combined with data-bases such as TCMSP and PubChem,active components were screened out and their corresponding targets were identified.These were then mapped against fracture disease targets obtained from databases like GeneCards to derive intersecting targets.Construction of"TCM-active ingredient-disease target"network and PPI network using Cytoscape 3.10.0 software and String database.Perform GO functional analysis and KEGG pathway enrichment analysis using the DAVID database.Finally,molecular docking technology was employed to validate the interaction between the active ingredient and the key target.Results:A total of 17 major active compo-nents of Sambucus adnata Wall.and their corresponding 357 target genes were identified,along with 2265 fracture disease target genes.The intersection yielded 127 target genes,from which 5 core active components and 12 core target genes were further screened out.The results of enrichment analysis showed that the key pathways of Sambucus adnata Wall.in the treatment of fracture were cAMP signaling pathway,cGMP-PKG signaling pathway,JAK-STAT signaling pathway and so on.The molecular docking results showed that the five core components of Sambucus adnata Wall.had high binding activities with key core targets.Conclusion:Sambucus adnata Wall.mainly treats fracture through the process of multi-component-multi-target-multi-pathway,mainly acting on the targets of MMP9,PTGS2,AKT1,ESR1,EGFR,etc,and through regulating the pathways of cAMP,cGMP-PKG,and JAK-STAT,it exerts the effects of analgesia and anti-inflammation,activating blood circulation and removing blood sta-sis,osteoclast proliferation and osteogenic differentiation,etc,so as to treat the fracture.This provides a theoretical basis for further research and clinical application of this drug.
