针对H5亚型禽流感病毒相对保守的3个区域,分别设计了 sgRNA和逆转录重组酶聚合酶恒温快速检测技术(RT-RPA)的特异性扩增引物,建立了基于RPA-CRISPR/Cas12b检测H5亚型禽流感病毒的方法,并对该检测方法的敏感性、特异性、重复性和可行性进行验证.结果显示,重组质粒阳性标准品检测的灵敏度达到了 1.0 copies/μL,是荧光RT-PCR灵敏度的10倍;该方法与H7亚型禽流感病毒、H9亚型禽流感病毒、新城疫病毒(NDV)和鸡传染性支气管炎病毒(IBV)的核酸均无交叉反应;用该方法检测50份临床模拟样品并与国标推荐的实时荧光RT-PCR进行比较,结果两种检测方法符合率达100%.上述结果表明,本研究建立的H5亚型禽流感病毒RPA-CRISPR/Cas12b检测方法可为口岸基层的快速筛检工作提供技术支撑.

Three relatively conserved regions of the H5 subtype avian influenza virus were selected for the design of sgRNAs and specific amplification primers for reverse transcription recombinase polymerase amplification(RT-RPA).A detection method based on RPA-CRISPR/Cas12b was established for the rapid identification of H5 subtype avian influenza virus.The sensitivity,specificity,repeatability,and feasibility of this method were systematically evaluated.The results showed that the detection limit of the recombinant plasmid positive standard was 1.0 copies/μL,representing a tenfold increase in sensitivity compared with conventional fluorescence RT-PCR.No cross-reactivity was observed with nucleic acids from H7 subtype avian influenza virus,H9 subtype avian influenza virus,Newcastle disease virus(NDV),or infectious bronchitis virus(IBV).When 50 simulated clinical samples were tested,a 100% concordance rate was achieved between this assay and the nationally recommended real-time RT-PCR method.These findings indicate that the RPA-CRISPR/Cas12b detection method established in this study provides a reliable and rapid on-site screening tool for frontline personnel at ports of entry.
