旨在制备非洲猪瘟病毒(ASFV)多表位抗原并评价其在疫苗中的应用.本研究设计了分子伴侣(TF16)与ASFV多表位抗原原核重组质粒,可溶性表达了非洲猪瘟病毒多表位重组蛋白(ASFV-pBT),重组蛋白纯化、鉴定后免疫小鼠并进行免疫原性评价,为非洲猪瘟亚单位疫苗研究提供试验基础.结果表明,ASFV-pBT可在大肠杆菌中可溶性表达,并被ASFV抗体阳性血清特异性识别.免疫小鼠血清中抗体效价可达1∶32 000,且ASFV-pBT可有效激活小鼠T淋巴细胞的增殖,CD4+和CD8+T淋巴细胞较阴性对照组显著提升(P<0.001),IFN-γ和IL-4细胞因子的表达也显著高于阴性对照组(P<0.001).通过体内、体外双重验证表明ASFV-pBT重组蛋白展现出良好的免疫原性,能够有效诱导机体产生显著的体液免疫和细胞免疫,为ASFV表位疫苗的研发提供了参考.

This study aimed to prepare multi-epitope antigens of African swine fever virus(ASFV)and evaluate its application in vaccine.We designed a prokaryotic recombinant plasmid expressing the molecular chaperone TF16 and ASFV multi-epitope antigen,which can solubly express ASFV-pBT multi-epi-tope recombinant protein.After purification and identification,mice were immunized and their immuno-genicity was evaluated,providing a test basis for the research of subunit vaccines against African swine fever.The results showed that ASFV-pBT can be solubly expressed in Escherichia coli and specifi-cally recognized by ASFV antibody positive serum.The ASFV-pBT immunized group achieved serum antibody titers up to 1∶32 000,and ASFV-pBT could effectively activate the proliferation of T lymphocytes in mice.Compared with the negative control group,mice in the ASFV-pBT group exhibited significantly in-creased proportions of CD4+and CD8+T lymphocytes(P<0.001)and IFN-γ and IL-4 cytokine levels were markedly elevated(P<0.001).In vivo and in vi tro double validation showed that ASFV-pBT recombinant protein possessed strong immunogenicity,inducing robust humoral and cellular immune respons es,thereby laying reference for ASFV epitope vaccine development.
