基于发泡聚丙烯载体的爪哇虫草MSC-F1固态发酵工艺优化OA
Optimization of Solid-state Fermentation Process for Cordyceps javanica MSC-F1 Based on Expanded Polypropylene Carrier
爪哇虫草Cordyceps javanica MSC-F1是一株对柑橘木虱Diaphorina citri和柑橘全爪螨Panonychus citri等多种害虫具有强致病力的生防真菌,但其产业化应用因发酵成本高和工艺不完善而受到限制.为了提高该菌株的产孢量并降低生产成本,本研究以惰性材料发泡聚丙烯粒子EPP为主要载体,以产孢量为指标,采用单因素和正交试验对培养基成分和发酵条件进行优化.结果表明,最佳基础培养基为EPP和稻壳混合物,比例为4∶1(V/V)、料水比1∶1.2.在此基础上,培养基中添加玉米粉20%、黄豆粉16%、酵母粉1.5%;固态发酵培养条件:培养温度25 ℃,培养时间9d,接种量2%和初始pH7.0.在此优化条件下,菌株MSC-F1产孢量较优化前提高了 6.28倍.本研究为爪哇虫草MSC-F1在发泡聚丙烯粒子惰性载体上的固态发酵提供了重要的理论依据.
Cordyceps javanica MSC-F1 is a biocontrol fungus with significant pathogenicity against various pests such as Diaphorina citri and Panonychus citri.However,its industrialized application is hindered by high fermentation cost and imperfect fermentation processes.To enhance spore yield and reduce production cost,this study employed expanded polypropylene particles(EPP)as the primary inert carrier and optimized medium composition and culture conditions using single-factor and orthogonal test for high spore yield.The results indicated that the optimal basal medium consisted of a mixture of EPP and rice husk at a ratio of 4∶1(V/V),with a solid-to-liquid ratio of 1∶1.2.Additionally,the basal medium was supplemented with 20%corn flour,16%soybean meal,and 1.5%yeast extract.The optimal solid-state fermentation conditions were:cultivation temperature at 25 ℃,fermentation period for 9 days,inoculation size of 2%,and initial pH of 7.0.Under these optimized conditions,the spore yield increased 6.28 times compared to pre-optimization level.This study provides critical theoretical support for solid-state fermentation of Cordyceps javanica using EPP as inert carrier.
