甘肃张掖、金昌地区犊牛腹泻致病菌的分离鉴定、致病性及耐药性分析OA
Isolation,Identification,Pathogenicity and Drug Resistance Analysis of Pathogenic Bacteria from Diarrhea Calves in Zhangye and Jinchang,Gansu Province
本研究旨在了解甘肃省张掖和金昌地区犊牛腹泻的主要致病菌种类及其致病性和耐药流行情况,为当地防治犊牛腹泻提供用药参考.本研究在张掖和金昌的6个犊牛养殖场采集了41份样品(27份粪便样,4份乳样,10份土壤样),通过细菌分离培养、革兰氏染色、生长特性鉴别和16S rRNA基因测序进行致病菌分离鉴定;采用致病性试验和K-B纸片法药敏试验检测致病菌的致病性和耐药性.结果表明,41份样本中共分离出4株致病菌,其中JC01株、JC02株、JC03株为大肠杆菌(Escherichia coli),JC04株为肺炎克雷伯菌(Klebsiella pneumoniae).同源性分析结果表明,JC01株与大肠杆菌U 5/41菌株亲缘关系最近,JC04株与肺炎克雷伯菌臭鼻亚种(Klebsiella pneumoniae subsp.ozaenae)标准菌株亲源关系最近.致病性研究表明,JC01株和JC04株均具有很强的致病性,可造成脾和空肠组织严重病理损伤.药敏试验结果表明,3株大肠杆菌对β-内酰胺类(88.89%)、大环内酯类(100%)、四环素类(100%)、林可酰胺类(100%)和磺胺类(100%)抗菌药物的耐药性较高;1株肺炎克雷伯菌对β-内酰胺类、氨基糖苷类、大环内酯类、四环素类、林可酰胺类和磺胺类抗菌药物完全耐药,仅对头孢西丁、氧氟沙星和诺氟沙星敏感.综上,甘肃张掖、金昌地区犊牛腹泻致病菌主要为大肠杆菌和肺炎克雷伯菌,二者均具有很强的致病性,且均存在耐药谱广、耐药率高的特征.
This study aimed to investigate the species,pathogenicity,and antibiotic resistance prevalence of the primary pathogenic bacteria from diarrhea calves in Zhangye and Jinchang regions of Gansu province,providing a reference for local prevention and treatment.A total of 41 samples(27 fecal samples,4 milk samples,10 soil samples)were collected from 6 cattle farms in Zhangye and Jinchang.Pathogenic bacteria were isolated and identified by bacterial culture,Gram staining,growth characteristic identification,and 16S rRNA gene sequencing.Pathogenicity tests and Kirby-Bauer(K-B)disk diffusion were utilized to confirm pathogenicity and antibiotic resistance.The results showed that 4 pathogenic strains were isolated from 41 samples,among which JC01,JC02,and JC03 were identified as Escherichia coli,and JC04 was identified as Klebsiella pneumoniae.Homology analysis indicated that JC01 was most closely re-lated to Escherichia coli strain U 5/41,while JC04 showed the highest homology with the standard strain Klebsiella pneumoniae subsp.ozaenae.Pathogenicity studies indicated that both JC01 and JC04 exhibited strong pathogenicity,causing severe pathological damage to the spleen and jejunum.Antibiotic susceptibility testing revealed that the 3 E.coli strains exhibited high resistance to β-lactams(88.89%),macrolides(100%),tetracyclines(100%),lincosamides(100%),and sulfonamides(100%).The K.pneumoniae strain was fully resistant to β-lactams,aminoglycosides,macro-lides,tetracyclines,lincosamides,and sulfonamides,remaining susceptible only to cefoxitin,ofloxacin,and norfloxa-cin.In conclusion,the primary pathogens causing diarrhea in calves in the Zhangye and Jinchang were E.coli and K.pneumoniae,both of which exhibited strong pathogenicity and broad-spectrum,high-level antibiotic resistance.
