马红球菌重组ABC转运蛋白的表达及其免疫效果评价OA
Expression of Recombinant ABC Transporter Protein from Rhodococcus equi and Evaluation of Its Immunological Effect
马红球菌(Rhodococcus equi,R.equi)为胞内寄生菌,主要可导致马驹发生化脓性支气管肺炎,给马产业造成重大经济损失.毒力相关脂蛋白A(Virulence-associated protein A,VapA)是R.equi致病的关键毒力因子,尽管研究证明用其免疫动物对R.equi感染具有一定抑制作用,但尚未查询到抗R.equi的商品化疫苗.生物信息学分析显示,R.equi的ABC转运蛋白可作为抵抗R.equi感染的潜在保护性抗原.本研究用原核表达的方法制备重组ABC转运蛋白(rABC),并将rABC、重组VapA蛋白(rVapA,阳性对照)和PBS(阴性对照)与弗氏佐剂乳化后分别免疫BALB/c小鼠,检测其免疫效果.结果显示,获得了符合预期分子量(62 kDa)且纯化的rABC;rABC和rVapA免疫后14 d试验小鼠开始产生特异性抗体;免疫后42 d血清IgG抗体的P/N值≥15.0,IgG1和IgG2a抗体效价分别为1∶409600和1∶102400,脾脏TNF-α阳性CD4+和CD8+T细胞比例均较阴性对照组显著升高(P<0.05);攻菌后7 d,rABC免疫组的脾细胞TNF-α及IL-6分泌量均较阴性对照组显著升高(P<0.05),且减轻了肺脏组织的病理变化.本研究结果提示,ABC转运蛋白能诱导小鼠产生较强的体液免疫和细胞免疫应答,并可减轻肺脏病理损伤,是R.equi亚单位疫苗开发的潜在候选抗原.
Rhodococcus equi(R.equi)is an intracellular parasitic bacterium that causes septic bronchopneumonia in foals,resulting in significant economic losses to the equine industry.Virulence-associated protein A(VapA)is a key virulence factor for R.equi,although studies have demonstrated that immunisation of animals with VapA has a pro-tective effect against R.equi infections,a commercial vaccine against R.equi has not yet been investigated.Bioinfor-matics analyses showed that the ABC transporter protein of R.equi could be a potential protective antigen against R.equi infection.In this study,recombinant ABC transporter protein(rABC)was prepared by prokaryotic expression,and BALB/c mice were immunised with rABC,recombinant VapA(rVapA,positive control)and PBS(negative con-trol)emulsified with Freund's adjuvant,respectively,to test their immunisation effects.The results showed that rABC with the expected molecular weight(62 kDa)and purified was obtained;specific antibodies began to be pro-duced 14 d after immunisation with rABC and rVapA;serum IgG antibody P/N values were≥15.0 at 42 d post-immunisation,IgG1 and IgG2a antibody potencies were 1∶409 600 and 1∶102 400,respectively,and the proportion of TNF-α-positive CD4+and CD8+T-cell in the spleen were significantly higher than those of the negative control group(P<0.05);7 d after infecting the bacteria,TNF-α and IL-6 secretion of splenocytes were significantly higher in rABC immunised group compared with those of the negative control group(P<0.05),and the histopathological changes in the lungs were attenuated.The results of this study suggest that ABC transporter proteins induce strong humoral and cellu-lar immune responses and attenuate lung pathological damage in mice,and are potential candidate antigens for R.equi subunit vaccine development.
