滩羊不同时期皮肤组织KRTAP1-1、KRTAP6-1和TCHH基因表达模式研究OA
Study on the Expression Pattern of KRTAP1-1,KRTAP6-1 and TCHH Genes in Skin Tissue of Tan Sheep Lambs at Different Periods
本试验旨在研究滩羊不同时期皮肤组织中KRTAP1-1、KRTAP6-1和TCHH基因的表达模式.利用实时荧光定量PCR(Quantitative real-time PCR,qRT-PCR)和蛋白免疫印迹(Western blot,WB)方法测定了KRTAP1-1、KRTAP6-1和TCHH基因mRNA及其编码蛋白在滩羊0(对照)、35和60日龄皮肤组织中的表达量.qRT-PCR检测结果显示,随着日龄的增长,滩羊皮肤组织中KRTAP1-1基因的表达量逐步上升,35日龄和60日龄的表达量极显著高于0日龄(P<0.01),且60日龄的表达量显著高于35日龄(P<0.05).随着日龄的增长,KRTAP6-1基因表达量也逐步上升,60日龄滩羊皮肤组织中的表达量极显著高于0和35日龄(P<0.01),但0日龄与35日龄间没有显著差异(P>0.05).随着日龄的增长,TCHH基因表达量逐步下降,0日龄滩羊皮肤组织中的表达量显著高于35日龄(P<0.05),极显著高于60日龄(P<0.01),35日龄的表达量极显著高于60日龄(P<0.01).WB检测结果显示,KAP1-1蛋白在35日龄滩羊皮肤组织中的表达量显著高于0日龄(P<0.05),并极显著高于60日龄(P<0.01);TCHH蛋白表达量随日龄增长逐步下降,0日龄的表达量极显著高于60日龄(P<0.01),35日龄的表达量显著高于60日龄(P<0.05),但0日龄和35日龄间没有显著差异(P>0.05).
To explore the expression pattern of KRTAP1-1,KRTAP6-1 and TCHH genes in the skin tissues of Tan sheep lambs at different periods,quantitative real-time PCR(qRT-PCR)and Western blot(WB)were employed to determine the expression level of KRTAP1-1,KRTAP6-1 and TCHH mRNA and their encoded proteins in the skin of Tan sheep at day 0(control group),35 and 60 of age.The results showed that the expression level of KRTAP1-1 gene was gradually increased with the growth of age,and its expression level at day 35 and 60 was extremely significantly higher than that at day 0(P<0.01),and the expression level at day 60 was significantly higher than that at day 35(P<0.05).With the growth of age,the expression level of KRTAP6-1 gene was also gradually increased,and its expression level at day 60 was extremely significantly higher than that at day 0 and 35(P<0.01),but there was no significant difference at day 0 and 35(P>0.05).Contrast to the other two genes,the expression level of TCHH gene was gradually decreased with the growth of age,and its expression level at day 0 was significantly higher than that at day 35(P<0.05),extremely signifi-cantly higher than that at day 60(P<0.01),and the expression level at day 35 was significantly higher than that at day 60(P<0.01).The WB results showed that the expression level of KAP1-1 protein in the skin tissue of Tan sheep at day 35 was significantly higher than that at day 0(P<0.05),and extremely significantly higher than that at day 60(P<0.01).With the growth of age,the expression level of TCHH protein was gradually decreased,and its protein expression level at day 0 was significantly higher than that at day 60(P<0.01),the expression level at day 35 was significantly higher than that at day 60(P<0.05),but there was no significant difference at day 0 and 35(P>0.05).
