本研究旨在通过针对不同羊毛纤维特征的皮肤转录组测序分析,筛选出调节毛囊发育的关键候选基因,为深入揭示驱动藏羊羊毛性状发育的分子机制提供重要依据.采集成年高原型藏羊和欧拉羊的皮肤组织,进行石蜡切片和苏木精-伊红染色(HE);采用Illumina NovaSeq平台进行高通量测序(PE150);利用DESeq2进行差异表达基因(DEGs)分析(|log2FC|>1,FDR<0.05),结合GO、KEGG数据库进行功能富集分析;并通过qRT-PCR对候选基因进行表达验证.毛囊形态学研究表明,初级毛囊具有较大的直径和完整的结构,而次级毛囊则表现出较小的直径且数量较多.此外,高原型藏羊的毛囊密度较低,毛囊间间距较大;相比之下,欧拉羊的毛囊密度较高,毛囊间间距较小.转录组测序结果显示,共鉴定出1065个DEGs,其中上调基因434个,下调基因631个.通过GO和KEGG功能富集分析发现,这些DEGs显著富集于Wnt信号通路、细胞外基质受体相互作用、PPAR信号通路、黏附、PI3K-Akt信号通路,其中THBS3、COL6A3、COL6A2、COL6A1、COL1A1、COL1A2、OVOL2、MYOC、FABP3、MMP1、KIT、FGF5、DKK1、MMP7、WNT7B和WNT16等关键基因在毛囊发育的调控中发挥重要作用.qRT-PCR结果显示,所选的7个DEGs的皮尔逊相关系数为0.77,P值为3.95×10-5,表明RNA-seq数据具有较高的可靠性.本研究筛选到的THBS3、COL6A3、COL6A2、COL6A1、COL1A1、COL1A2、OVOL2、MYOC、FABP3、MMP1、KIT、FGF5、DKK1、MMP7、WNT7B和WNT16等关键基因表现出显著的差异表达模式,为揭示藏羊毛囊发育的分子机制提供了重要理论基础.

This study aims to identify key candidate genes regulating hair follicle development through transcriptome sequencing analysis of skin tissues associated with different wool fiber characteristics,thereby provding a crucial theo-retical basis for an in-depth elucidation of molecular mechanisms underlying wool trait development in Tibetan sheep.Skin tissues from adult plateau-type Tibetan sheep and Oula sheep were selected for paraffin sectioning and HE stain-ing.High-throughput sequencing(PE150)was conducted using the Illumina NovaSeq platform.Differential expression genes(DEGs)were analyzed using DESeq2(|log2FC|>1,FDR<0.05),and functional enrichment analysis was per-formed in combination with the GO and KEGG databases.The expression of candidate genes was verified by qRT-PCR.Morphological studies on hair follicle indicated that primary hair follicles possess a larger diameter and a com-plete structure,while secondary hair follicles are characterized by a smaller diameter and a higher quantity.Addition-ally,the hair follicle density of plateau-type Tibetan sheep was relatively low,with a larger distance between hair fol-licles;in contrast,Oula sheep exhibit a higher hair follicle density,with a smaller distance between hair follicles.Tran-scriptome sequencing results showed that a total of 1 065 DEGs were identified,including 434 up-regulated genes and 631 down-regulated genes.Through GO and KEGG functional enrichment analysis,it was found that these DEGs were significantly enriched in the Wnt signaling pathway,extracellular matrix receptor interaction,PPAR signaling pathway,adhesion,and PI3K-Akt signaling pathway.Among them,key genes such as THBS3,COL6A3,COL6A2,COL6A1,COL1A1,COL1A2,OVOL2,MYOC,FABP3,MMP1,KIT,FGF5,DKK1,MMP7,WNT7B and WNT16 played important roles in the regulation of hair follicle development.The qRT-PCR results showed that the Pearson correlation coeffi-cient of the selected 7 DEGs was 0.77,and the P value was 3.95×10-5,indicating that the RNA-seq data had high reli-ability.The identified key genes including THBS3,COL6A3,COL6A2,COL6A,WNT7B and WNT16 exhibited significant differential expression patterns,providing an important theoretical basis for revealing the molecular mechanism of hair follicle development in Tibetan sheep.
