马铃薯StVac14基因功能的初步解析OA
Preliminary Analysis of the Function of StVac14 Gene in Solanum tuberosum L.
干旱是影响马铃薯生长发育的重要因素,Vac14基因在植物生长发育与稳态调控过程中发挥重要作用.通过对马铃薯栽培种Vac14同源蛋白进行比对分析,发现马铃薯、番茄和辣椒中Vac14蛋白较为保守;表达量分析发现,StVac14在7个不同组织中均表达,且在干旱胁迫下显著上调;将StVac14基因敲除后,植株耐旱性显著降低,脯氨酸积累量显著下降,丙二醛含量增加.明确了 StVac14在马铃薯响应干旱胁迫中的正调控作用,为深入解析StVac14基因响应马铃薯干旱胁迫的机制奠定了基础.
Drought is a critical factor influencing the growth and development of potato(Solanum tu-berosum L.).The Vac14 gene plays a pivotal role in plant growth,development,and homeostasis regu-lation.In this study,comparative analysis of Vac14 homologous proteins in cultivated potato varieties demonstrated that Vac14 proteins are highly conserved among potato,tomato(Solanum lycopersi-cum),and pepper(Capsicum annuum).Expression level analysis revealed that StVac14 is ubiquitously expressed in seven different tissues and is significantly upregulated under drought stress.Following StVac14 gene knockout,the drought tolerance of the plants was remarkably reduced,the accumulation of proline was significantly decreased,and the content of malondialdehyde(MDA)was increased.This study clarifies the positive regulatory role of StVac14 in potato's response to drought stress,laying a solid foundation for further elucidating the mechanism by which the StVac14 gene mediates potato's response to drought stress.
