Identification of Key Genes Underlying Freezing Tolerance in the Wild Potato Solanum commersonii

This study aimed to elucidate the molecular mechanisms underlying freezing tolerance in potato.U-sing the cold-tolerant wild species Solanum commersonii as material, transcriptome sequencing was performed after a 2-hour treatment at-4℃,resulting in the identification of 4 588 differentially expressed genes, among which upregulated genes accounted for 55.70%.Functional analysis revealed that low-temperature stress sig-nificantly activated freezing resistance-related pathways such as starch and sucrose metabolism,glycolysis/glu-coneogenesis,while inhibiting growth-related metabolism including photosynthesis.Further investigation iden-tified 18 key genes that were significantly upregulated under low temperature, primarily involved in sucrose metabolism, which coordinately regulate the synthesis and breakdown of carbohydrates to provide cellular pro-tection and energy. These findings reveal the molecular basis by which S.commersonii adapts to low tempera-ture through metabolic reprogramming, offering crucial candidate genes and theoretical support for improving freezing tolerance in cultivated potato.
