游离circCUX1在卵巢癌患者血清中的表达及诊断价值研究OA
Expression and diagnostic value of serum-free circCUX1 in ovarian cancer patients
目的:探索游离环状RNA circCUX1 在卵巢癌患者血清中的表达水平及其临床诊断价值.方法:利用DNA琼脂糖凝胶电泳、Sanger测序、放线菌素D实验和RNase R实验对circCUX1 的分子特征进行分析.选取2024 年12 月至2025 年10 月在山东大学齐鲁医院诊治的30 例卵巢癌患者和30 例女性健康志愿者.RT-qPCR法检测患者血清circCUX1水平.采用受试者工作特征(ROC)曲线评估circCUX1 对卵巢癌的诊断效能.结果:circ-CUX1 由CUX1 基因的外显子反向剪接环化形成.与CUX1 mRNA相比,circCUX1 在卵巢癌细胞中表现出更高的稳定性.与对照组相比,卵巢癌组血清游离circCUX1 水平升高(P<0.05).卵巢癌患者中Ⅲ/Ⅳ期血清游离circCUX1 水平高于Ⅰ/Ⅱ期(P<0.05).发生远处转移患者血清游离circCUX1 水平高于未发生远处转移患者(P<0.05).血清游离circCUX1 水平与FIGO分期、淋巴结转移和远处转移相关(均P<0.05).ROC曲线显示,血清CA125、circ-CUX1 单独及联合诊断卵巢癌的曲线下面积(AUC)分别为0.786、0.867、0.928,联合诊断与CA125 单独诊断之间的差异有统计学意义(P<0.05).结论:卵巢癌患者血清中游离circ-CUX1 水平升高,且其表达水平随疾病进展逐渐增加,提示circCUX1 对卵巢癌的辅助诊断具有一定的临床应用价值.
Objective:To explore the expression levels of serum-free circCUX1 in ovarian cancer patients and to evaluate its clinical diagnostic value.Methods:The molecular characteris-tics of circCUX1 were analyzed using DNA agarose gel electrophoresis,Sanger sequencing,actino-mycin D treatment,and RNase R digestion assays.A total of 30 patients diagnosed with ovarian cancer between December 2024 and October 2025 at Qilu Hospital of Shandong University were enrolled as the ovarian cancer group,along with 30 healthy volunteers as the control group.Serum levels of circCUX1 were quantified using reverse transcription-quantitative polymerase chain reac-tion(RT-qPCR).Receiver operating characteristic(ROC)curve analysis was performed to eval-uate the diagnostic efficacy of serum circCUX1 for ovarian cancer.Results:CircCUX1 was genera-ted from the CUX1 gene through back-splicing of exons.Compared with linear CUX1 mRNA,circ-CUX1 exhibited greater stability in ovarian cancer cells.Serum-free circCUX1 levels were signifi-cantly elevated in ovarian cancer patients compared with healthy controls(P<0.05).Moreover,serum cell-free circCUX1 levels were higher in patients with advanced-stage disease(FIGO sta-gesⅢ/Ⅳ)than in those with early-stage disease(FIGO stagesⅠ/Ⅱ)(P<0.05),and were also increased in patients with distant metastasis compared with those without distant metastasis(P<0.05).Serum-free circCUX1 levels were significantly associated with FIGO stage,lymph node metastasis,and distant metastasis(all P<0.05).Receiver operating characteristic(ROC)curve analysis demonstrated that the areas under the curve(AUC)for serum CA125,circCUX1,and their combination in diagnosing ovarian cancer were 0.786,0.867,and 0.928,respectively.The combined diagnostic model showed a statistically significant improvement compared with CA125 alone(Z=2.13,P<0.05).Conclusion:circCUX1 is highly expressed in the serum of ovarian cancer patients,and its level increases with disease progression,indicating that circCUX1 may have potential clinical value as an auxiliary diagnostic biomarker for ovarian cancer.
