利用EST-SSR分子标记评价古槐群体的遗传多样性,并解析其群体结构以及群体之间的亲缘关系,为国槐优质种质资源筛选及古树保护工作提供科学借鉴.16对EST-SSR分子标记评价来自6个群体91份供试材料的遗传多样性,通过遗传参数评价古槐群体的遗传多样性丰富度;采用AMOVA方差分析探究群体的遗传分化水平;STRUCTURE群体结构分析、PCoA主坐标分析以及UPGMA聚类分析互相验证揭示群体的遗传结构.结果表明,EST-SSR分子标记共扩增出了90个等位基因,表现出较丰富的遗传多样性水平(I=1.056、H=0.572、He=0.571).AMOVA分析表明,95%的遗传变异发生在群体内部.群体间的遗传分化系数Fst均值为0.046,遗传分化程度处于较低水平.STRUCTURE分析、PCoA分析以及UPGMA分析的结果较为一致,6个群体可分为3个亚群.综上所述,供试古槐群体遗传多样性水平较丰富,群体间存在着持续且有效的基因交流,整体上未表现明显的近交衰退.山东潍坊(WF)古槐群体表现最佳,应充分保护并合理利用;北京(BJ)和山东淄博(ZB)古树群体存在杂合子略微缺失、遗传多样性低等问题,应及时保育.

This study employed EST-SSR molecular markers to evaluate the genetic diversity of ancient So-phora japonica populations,analyze their population structure,and elucidate genetic relationships among them.The aim is to provide a scientific basis for selecting high-quality germplasm and to inform conserva-tion strategies for ancient trees.Genetic diversity was assessed using 16 EST-SSR molecular markers across 91 samples from six populations.AMOVA was performed to determine the level of genetic differen-tiation,while STRUCTURE,PCoA and UPGMA analyses were used to reveal the genetic structure of pop-ulations.Results indicated that the EST-SSR molecular markers amplified a total of 90 alleles,demonstra-ting a moderate to high level of genetic diversity(I=1.056,H=0.572,He=0.571).AMOVA revealed that 95%of genetic variation occurred within populations.The mean interpopulation genetic differentiation coefficient Fst was 0.046,indicating low genetic differentiation.Consistently,the STRUCTURE,PCoA,and UPGMA analyses grouped the six populations into three distinct subpopulations.In summary,the studied ancient Sophora japonica populations exhibit relatively high genetic diversity.Sustained and effective gene flow exists between them,and no significant inbreeding depression was observed overall.The WF popula-tion shows the highest genetic diversity and warrants full protection and rational utilization;the BJ and ZB populations exhibit a slight heterozygote deficiency and lower genetic diversity,necessitating timely conser-vation efforts.
