苦豆碱通过抑制cGAS-STING通路减轻顺铂诱导的急性肾损伤的机制研究OA
Aloperine ameliorates cisplatin-induced acute kidney injury by inhibiting the cGAS-STING pathway
目的 阐明苦豆碱通过环磷酸鸟苷-腺苷合成酶(cGAS)-干扰素基因刺激因子(STING)通路发挥肾脏保护的作用机制.方法 雄性8周SPF级C57小鼠18只,随机分成对照组,模型组,苦豆碱组.除对照组外,均通过腹腔注射顺铂来建立小鼠急性肾损伤模型,持续给药3d后处死.取肾脏组织进行HE切片染色计算损伤评分,并进行流式细胞术检测评估小鼠肾脏免疫细胞浸润和活化程度,同时取肾脏组织进行Western blot和qPCR检测,用免疫荧光测定IRF3细胞内分布情况.结果 与对照组比较,模型组的肾脏损伤评分显著升高(P<0.0001),炎性巨噬细胞浸润明显增加,IL-6,IFNβ,CXCL10,cGAS,STING的mRNA表达水平均显著上升(P<0.05,P<0.01,P<0.001,P<0.0001),TBK1,IRF3蛋白磷酸化形式表达升高(P<0.001,P<0.0001),IRF3蛋白入核增加(P<0.0001).与模型组比较,苦豆碱给药组肾脏损伤评分,巨噬细胞浸润,炎症细胞因子转录,STING蛋白,TBK1,IRF3蛋白磷酸化均出现不同程度的降低.结论 苦豆碱作为STING通路的天然拮抗剂对顺铂诱导的AKI具有保护作用.
Objective To elucidate the nephroprotective mechanism of aloperine via the cyclic GMP-AMP synthase(cGAS)-stimulator of interferon genes(STING)pathway.Methods Eighteen 8-week-old male specific pathogen-free(SPF)C57BL/6 mice were randomly assigned to control,model,and aloperine-treated groups.Except for the control group,acute kidney injury(AKI)was induced by a single intraperitoneal injection of cisplatin.After 3 days of continuous drug administration,mice were euthanized.Kidney tissues were collected for HE staining to assess histological injury scores.Flow cytometry(FCW)was employed to evaluate renal immune cell infiltra-tion and activation.Western blot(WB)and quantitative real-time polymerase chain reaction(qPCR)were performed on kidney homog-enates.Immunofluorescence(IF)was conducted to determine the subcellular localization of IRF3.Results Compared with the control group,the model group exhibited a significantly higher kidney injury score(P<0.0001)and increased infiltration of inflammatory mac-rophages.The mRNA levels of IL-6,IFN-β,CXCL10,cGAS,and STING were significantly upregulated in the model group(all P<0.05).The phosphorylation levels of TBK1 and IRF3 proteins were elevated(P<0.001 and P<0.0001,respectively),accompanied by enhanced nuclear translocation of IRF3(P<0.0001).Compared with the model group,aloperine treatment significantly attenuated the cisplatin-induced increases in kidney injury score,macrophage infiltration,pro-inflammatory cytokine transcription,expression of STING protein,and phosphorylation of TBK1 and IRF3.Conclusion Aloperine alleviates cisplatin-induced acute kidney injury,poten-tially by functioning as a natural antagonist of the STING signaling pathway.
